The present invention provides a method for generation of multi-domain proteins, more particular antibodies, with an improved native state colloidal stability. The present disclosure provides a method of producing an IgG1 or IgG4 antibody with an improved colloidal stability comprising, in embodiments: calculating for each domain of said IgG1 or IgG4 antibody the total net charge at a given pH; introducing one or more modifications to the amino acid residues of the constant region of said IgG1 or IgG4 antibody to minimize the charge sign difference between the domains, wherein said one or more modification is, or each are, a substitution of a charged amino acid by a polar (non-charged) amino acid; and producing the modified multi-domain protein with improved colloidal stability at the given pH.

Multi-domain proteins with increased native state colloidal stability

This invention is in the area of improved anti-aP2 antibodies and antigen binding agents, and compositions thereof, which target the lipid chaperone aP2/FABP4 (referred to as 'aP2') for use in treating disorders such as diabetes, obesity, cardiovascular disease, fatty liver disease, and/or cancer, among others. In one aspect, improved treatments for aP2 mediated disorders are disclosed in which serum aP2 is targeted and the biological activity of aP2 is neutralized or modulated using low-binding affinity aP2 monoclonal antibodies, providing lower fasting blood glucose levels, improved systemic glucose metabolism, increased systemic insulin sensitivity, reduced fat mass, reduced liver steatosis, reduced cardiovascular disease and/or a reduced risk of developing cardiovascular disease.

Anti-AP2 antibodies and antigen binding agents to treat metabolic disorders

The present invention provides an antibody of the class IgG4 comprising at least one heavy chain which comprises a C1 domain and a hinge region, wherein in each heavy chain: a. the inter-chain cysteine at position 127, numbered according to the Kabat numbering system, in the C1 domain is substituted with another amino acid; and b. one or more of the amino acids positioned in the upper hinge region is substituted with cysteine.

In one aspect, improved or novel treatments for IgE mediated disorders are disclosed in which IgE is targeted (for instance free IgE and/or IgE complexed with the FcεRI receptor).
