Nortriptyline functionalized compounds are provided for conjugation to antigenic compositions, particularly poly(amino acids), and enzymes. The antigenic conjugates are employed for the production of antibodies, which find use in immunoassays for the determination of nortriptyline in a sample, while the enzyme conjugates find use in an enzyme assay for the determination of nortriptyline in a sample.

Methods and devices are provided for carrying out assays with minimal technical training where quantitative visual determinations may be made. The devices provide for a flow-path where a sample receiving element is moved from a sample receiving position to a position where it serves to bridge a transport element and a measurement element, so that the sample may be transported from the sample element to the measurement region. Labelled conjugates are provided which migrate a distance into the measurement region related to the amount of analyte in the sample.

Systems are provided for the detection of hydrogen peroxide for determination of analytes, where a N-alkylated 3-oxyaniline derivative is covalently bonded to a bibulous support and the other component for forming an indamine dye may be non-covalently bound to the support. Upon passage of a transport medium across the support in the presence of hydrogen peroxide and a peroxidase, an indamine dye is formed. Where a limited amount of hydrogen peroxide is introduced at a site proximal to one end of the support, and the hydrogen peroxide migrates toward the other end, the distance of color development will be related to the amount of hydrogen peroxide, which can be correlated to the amount of analyte in a sample.

A self-contained assay device is provided, where an analyte may be detected or be quantitated by measuring the distance of a detectable signal from a predetermined site. The device employs two strips separated by a gap, where a flow path is completed by movement of a sample receiving pad into the gap. Various chemistries may be employed for providing a detectable signal.

A method for detecting boar taint in a pig sample is provided. Anti-skatole antibodies are combined in a reaction buffer with a pig sample to form a reaction mixture. After incubating, the amount of skatole-antibody complex formation is determined. In a particular embodiment, the method utilizes solid substrate-affixed antibodies in a competitive inhibition assay with an enzyme-labeled skatole analogue. Kits facilitating the method are also provided.

Chloramphenicol derivatives are provided for use in the preparing of antigen conjugates for the production of antibodies specifically for chloramphenicol. Specifically, the aryl amino group is derivatized to introduce a non-oxo-carbonyl group which is used for amide formation with an antigen. The conjugate is then injected into a vertebrate for production of antisera which is isolated in conventional ways and find particular use in competitive protein binding assays.

In particular, the compounds of the present invention can be used for measuring the presence of a .beta.-lactamase in a patient serum sample by adding a known amount of penicillin to the sample and observing the production of penicilloic acid over a fixed period of time.
