The present invention relates to a method for constructing a host cell expressing a polypeptide of interest from at least two ORF's stably integrated onto the chromosome of the host cell comprising the steps of:

Stably maintained multiple copies of at least two ORF in the same orientation

The invention provides methods of screening a microbial host cell for a property of interest in a microfluidic device, the method comprising the steps of: a) transforming a d-alanine racemase-deficient microbial host cell with a polynucleotide construct comprising: i) one or more polynucleotide region providing the property of interest when present in the host cell, and ii) at least one polynucleotide region complementing the d-alanine racemase deficiency when present in the host cell; and b) screening the transformed host cell for the property of interest in the microfluidic device in the absence of externally provided d-alanine.

Microfluidic device screening method

The present invention relates to novel JP170 like subtilases from wild-type bacteria, hybrids thereof and to methods of construction and production of these proteases. Further, the present invention relates to use of the claimed subtilases in detergents, such as a laundry or an automatic dishwashing detergent.

Subtilases

The invention relates to the use of co-expression of an antimicrobial agent and an enzyme, with a view to improving yield and/or overall production economy. Examples of antimicrobial agents are antimicrobial peptides such as lactoferricins and antimicrobial enzymes such as lysozyme and glucose oxidase, and examples of enzymes are endoglucanase, xylanase, phytase, protease, galactanase, mannanase, dextranase, alpha-galactosidase, pectate lyase, alpha-amylase and glucoamylase. A fusion product comprising the antimicrobial agent and the enzyme and a cleavable linker is novel, and can be used in animal feed and animal feed additives. The invention also describes the use of a protection domain wherein at least 50% of the amino acid residues comprised in the peptide protection domain are D (Asp) and/or E (Glu). The protection or quenching domain serves to temporarily and reversibly inactivate the antimicrobial peptide during the expression.

Recombinant production of antimicrobial peptides

The present invention relates to subtilases having a reduced tendency towards inhibition by substances present in eggs, such as the trypsin inhibitor type IV-0. These subtilases are useful exhibiting excellent or improved wash performance on egg stains when used in e.g. cleaning or detergent compositions, such as laundry detergent compositions and dishwash compositions, including automatic dishwash compositions.

The present invention relates to isolated nucleic acid sequences encoding polypeptides having proteolytic activity. The invention also relates to nucleic acid constructs, vectors, and host cells comprising the nucleic acid sequences as well as recombinant methods for producing the polypeptides.
