The present invention relates to: a newly developed triazine-cored amphiphilic compound; a preparation method therefor; and a method for extracting, solubilizing, stabilizing, crystallizing or analyzing membrane proteins by using the same. In addition, the compound enables membrane proteins, which have various structures and characteristics, to be efficiently extracted from cell membranes and stably stored in an aqueous solution for a long time, compared to a conventional compound, thereby being usable in functional and structural analysis thereof. Analyzing the structure and function of membrane proteins is closely related to the development of a novel drug, and thus is one of the greatest interests in the biology and chemistry fields.

The present invention relates to: amphipathic compounds derived from 1,3-acetonedicarboxylate; a preparation method therefor; and a method for extracting, solubilizing, stabilizing, crystallizing or analyzing membrane proteins by using the same. The present invention relates to a vitamin E-based amphipathic compound, a method for producing same, and a method for extracting, solubilizing, stabilizing, or crystallizing a membrane protein using same. By using a compound according to the present invention, not only is an excellent membrane protein extraction and solubilization effect achieved, but the membrane protein can be stably stored for a long period of time in an aqueous solution, and thus the compound can be utilized in analyzing the function and structure of the membrane protein. Moreover, the vitamin E-based amphipathic compounds exhibited superb properties in the visualization of protein compounds through an electron microscope. Membrane protein structure and function analysis is one of the fields of greatest interest in biology and chemistry today, and since at least half of new drugs currently being developed target membrane proteins, the vitamin E-based amphipathic compounds may be applied to the membrane protein structure research, which is closely related to the development of new drugs.

The present invention relates to an amphiphilic compound having a dendronic hydrophobic group, a method for preparing the same, and a method for extraction, solubilization, stabilization, or crystallization of a membrane protein by using the same. The use of the compound according to the present invention leads to an excellent membrane protein solubilization effect and a stable storage of a membrane protein in an aqueous solution for a long time, and thus can be utilized for functional analysis and structural analysis of the membrane protein. Especially, the amphiphilic compound having a dendronic hydrophobic group showed very remarkable characteristics in the visualization of protein composites through an electronic microscope. Provided are a newly developed norbornene-based amphiphilic compound, a method for preparing the same, and a method for extracting, solubilizing, stabilizing, crystallizing or analyzing a membrane protein using the same. In addition, the compound may effectively extract membrane proteins having various structures and characteristics, compared to a conventional compound, from cell membranes, and may be stably stored in an aqueous solution for a long period of time, and therefore may be used in their functional analysis and structural analysis. The structural and functional analyses of membrane proteins are the most noticeable field in biology and chemistry today due to a close relationship to the development of new drugs.

Norbornene-based amphipathic compound and utilization thereof

A newly developed butane-tetraol-based amphiphilic compound, a method of preparing the same, and a method of extracting, solubilizing, stabilizing, crystallizing or analyzing a membrane protein using the amphiphilic compound are provided. The butane-tetraol-based compound is found to have a central structure exhibiting chirality, and isomers of the compound have clearly different characteristics according to the stereochemistry of the central structure, thereby making it possible to select compounds suitable for the uses thereof. The present invention relates to a mannitol-based amphipathic compound, a method of preparing the same, a method of extracting, solubilizing, stabilizing or crystallizing a membrane protein using the compound, and a method of analyzing a structure of the membrane protein under an electron microscope using the compound. When the mannitol-based compound according to the present invention is used, the membrane protein can be stably stored in an aqueous solution for a prolonged period of time and thus can be applied to analysis of functions and structures thereof. The present invention relates to a TRIS- or neopentyl glycol-based amphipathic compound, a method of preparing the same, and a method of extracting, solubilizing, stabilizing, or crystallizing a membrane protein using the same. The present invention relates to a xylene-based amphiphilic compound, a method for preparing the same, and a method for extracting, solubilizing, stabilizing or crystallizing a membrane protein using the same. By using the xylene-based compound according to the present invention, a membrane protein may be stably stored in an aqueous solution for a long time, and may be applied in functional and structural analyses. The structural and functional analysis of the membrane protein is one of the fields of highest interest in biology and chemistry, and the xylene-based compound according to the present invention can be applied in research on protein structure that is closely related to development of a new drug.

Mesitylene-cored amphiphiles and uses thereof

The invention provides tandem facial amphiphiles for biochemical manipulations and characterization of membrane proteins, such as intrinsic membrane proteins. Members of this new family display favorable behavior with several membrane proteins. These amphiphiles can form relatively small micelles, and small changes in amphiphile chemical structures can result in large changes in their physical properties. The tandem facial amphiphiles can be used to aid the solubilization, isolation, purification, stabilization, crystallization, and/or structural determination of membrane proteins.

Bringing membrane proteins into aqueous solution generally requires the use of detergents or other amphiphilic agents. The invention provides a new class of amphiphiles, each of which includes a multi-fused ring system as a lipophilic group. These new amphiphiles confer enhanced stability to a range of membrane proteins in solution relative to conventional detergents, leading to improved structural and functional stability of membrane proteins, including integral membrane proteins. Accordingly, the invention provides new amphiphiles for biochemical manipulations and characterization of membrane proteins. These amphiphiles display favorable behavior with membrane proteins and can be used to aid the solubilization, isolation, purification, stabilization, crystallization, and/or structural determination of membrane proteins.

Amphiphilic compounds

The invention provides amphiphilic compounds and methods for manipulating membrane proteins. Compounds of the invention, for example, the compounds of Formulas I-XIX, can be prepared from readily available starting materials. The amphiphilic compounds can manipulate membrane protein at relatively low concentrations compared to many known detergents. The compounds can be used to aid the isolation of membrane proteins, for example, to aid their solubilization and/or purification. The compounds can also be used to aid the functional and structural determination of membrane proteins, including their stabilization and crystallization.
