Biological sample processing system comprising an imaging unit () comprising a digital image processing system and at least one microscope including at least one lens (), a sample processing station () comprising a handling platform () including a support () and a displacement mechanism for moving the support (), and a sample processing unit () mounted on the handling platform (). The sample processing unit () comprises a tissue slide holder () for mounting thereon a tissue slide () with a biological sample () fixed thereon and a microfluidic cartridge holder () for mounting a microfluidic cartridge () thereon. The tissue slide holder () is coupled to the microfluidic cartridge holder () via a coupling () allowing the microfluidic cartridge and the tissue support to be mounted and removed from the sample processing unit in an opened position, and in a closed position for the tissue support () to be in sealing contact with the microfluidic cartridge (). The sample processing station comprises a plurality of said sample processing units mounted on the handling platform () and moveable from a position allowing mounting of the tissue slide, respectively microfluidic cartridge, or removal thereof, to a position in which the viewing window in the microfluidic cartridge holder () is positioned in alignment with the lens of said at least one microscope.

Biological sample processing system and microfluidic cartridge therefor

Microfluidic network device () configured to supply reagents to a biological tissue sampling device (), comprising a plurality of microfluidic inlet channels () connected to respective sources of said reagents, at least one common outlet channel (), and a plurality of valves () interconnecting an outlet end () of each of said plurality of inlet channels to said at least one common outlet channel.

Microfluidic network device

Microfluidic cartridge () comprising a sampling device () having a sealing ring () arranged to form a microfluidic chamber () when a support containing a biological sample is brought into contact with the sealing ring, and a microfluidic network device () configured to supply reagents to the microfluidic chamber. The sampling device further comprises inlet and outlet distribution networks () in fluid communication with the microfluidic chamber and a slide holder () to guide and position said support containing a biological sample on the sampling device. The microfluidic network device comprises a plurality of reagent inlet channels () fluidly connectable to reagent sources, at least one reagent outlet channel () fluidly connected to the sampling device inlet distribution network (), and a plurality of valves () operable to selectively connect the inlet channels to the at least one outlet channel. The sampling device () and microfluidic network device () are formed on a common microfluidic support () as a single part.

The invention relates to a method for in situ imaging of samples by cycle multiplexing that enables imaging of various molecular targets through multi-molecular read-outs on the same sample in a rapid, highly sensitive and reliable manner. The invention is further related to imaging buffers preventing the degradation of the sample and of the imaging reagents, which are particularly useful in a method of in situ imaging of samples by cycle multiplexing according to the invention.
