The present disclosure provides compositions and methods for regulating expression of transcribable polynucleotides in plant cells, plant tissues, and plants. Compositions include regulatory polynucleotide molecules capable of providing expression in plant tissues and plants. Methods for expressing polynucleotides in a plant cell, plant tissue, or plants using the regulatory polynucleotide molecules disclosed herein are also provided.

Regulatory polynucleotides and uses thereof

A plant growth array device includes an aerial growth chamber configured to receive aerial shoot portions of a plurality of plants and a root growth chamber configured to receive root portions of the plurality of plants. A dividing member is between the aerial growth chamber and the root chamber and has a plurality of apertures for receiving the plurality of plants therein. The plurality of apertures are configured so that the root portions grow substantially in a common orientation.

The homeotic gene AGL42 inhas been cloned and characterized and is presented herein, both alone and in association with expression control sequences and in vectors which may be used e.g. for transgenic expression. The gene exhibits the role of controlling meristem maintenance. A labeled construct is shown to serve as a marker for identity of the quiescent cells within the meristem. Both the DNA and protein are disclosed.

Homeotic genes and proteins, uses thereof

The present invention provides compositions and methods for regulating expression of heterologous nucleotide sequences in a plant. Compositions include a novel nucleotide sequence for a root-preferred promoter for the gene encoding CRWAQ81. A method for expressing a heterologous nucleotide sequence in a plant using the promoter sequences disclosed herein is provided. The method comprises stably incorporating into the genome of a plant cell a nucleotide sequence operably linked to the root-preferred promoter of the present invention and regenerating a stably transformed plant that expresses the nucleotide sequence.

A method by which many plants control organ shape is by regulated, differential cellular expansion. A gene involved in regulating the expansion of plants cells, such asroot cells is COBRA which encodes a protein with a putative GPI anchor. Plants comprising altered root morphologies may be produced by control of COBRA activity. For example, roots which lack COBRA activity comprise thicker, fatter roots (a CORE or cob phenotype) which are well suited for penetrating dense, compacted soil. Plants lacking COBRA activity may be useful in applications wherein plant growth in areas with dense soil would be beneficial. The present invention comprises theCOBRA gene and COBRA protein and homologues thereof as well as mutated alleles of COBRA. Also provided are anti-COBRA antibodies, transgenic plants which overexpress COBRA and methods for identifying COBRA modulating substances.
