The present invention provides methods for constructing peptide construct sets and methods of use of these peptide construct sets in assay systems for peptide analysis, and in particular for use in high throughput peptide analysis. The methods allow for analysis of large sets of peptide constructs in a cost-effective manner, employing molecular biological techniques that are both robust and easily parallelized. Thus, the methods allow for the construction of peptide construct sets encompassing, e.g., the human proteome.

Peptide constructs and assay systems

The present invention provides devices and methods for detecting and capturing molecular biomarkers from a subject in situ. Specifically, the devices contain an array of microneedles to which are attached probes specific for one or more biomarkers of interest. The devices can be used directly on a subject (e.g., via skin piercing) in detecting the biomarkers in the body of the subject (e.g., tissues, blood stream).

Methods and devices for detection and acquisition of biomarkers

Embodiments of present application are directed to microfluidic devices and particularly digital micro-plastic fluidic devices that are specifically designed to prevent sample contamination during sample processing, methods of manufacturing the same, and methods to improve sample analysis process by preventing sample contamination.

The present disclosure generally relates to nucleic acid amplification systems and methods suitable for construction of nucleic acid samples, including construction of normalized nucleic acid libraries. In some embodiments, the method includes providing one or more input nucleic acid samples, contacting each of the input nucleic acid samples (e.g., input library) with a reaction mixture including first amplification or normalization primers and second amplification or normalization primers, wherein the first amplification or normalization primers are immobilized on a solid support and the second amplification or normalization primers are in solution phase, and amplifying the input nucleic acid samples under conditions such that substantially all of the first amplification or normalization primers are incorporated into amplification products. Further provided are systems and droplet actuator devices that are configured to carry out the methods disclosed herein. Compositions that include nucleic acid samples and libraries, preferably normalized, prepared in accordance with the disclosed methods and systems are also provided.

Methods and systems for construction of normalized nucleic acid libraries

The present disclosure provides polynucleotide conjugates, methods, and assay systems for use in detecting the presence, absence, and/or amount of an analyte in a sample. Various polynucleotide conjugates, conjugate pairs, sets, libraries, and assay systems comprising the same are disclosed. In particular, methods and assay systems for antibody detection and analysis are provided. For example, assays capable of high levels of multiplexing are used for antibody detection and analysis in a biological sample, e.g., Lyme disease patient samples. The presently disclosed polynucleotide conjugates, methods, and assay systems can be used to provide sensitive and reliable diagnosis, even at early stages of a disease or condition. Use for monitoring disease progression and prognosis is also disclosed.
