Media, kits, and methods for cultivation of mammalian embryos from fertilized oocyte stage to blastocyst stage in the presence of N-benzyl-3,5-dicarbethoxy-4-phenyl-1,4-dihydropyridine. The use of this supplement improves the early embryonic development, in particular the quality of zygotes and the blastocyst formation. The supplemented media can be used with pigs, humans, or other mammals N-benzyl-3,5-dicarbethoxy-4-phenyl-1,4-dihydropyridine may be added to known media, and used for with vitro fertilization methods.

Supplement for cultivation of mammalian embryos

The present invention provides a new and improved sperm stimulating additive comprising a certain amount of inorganic pyrophosphate (PPi). Addition of PPi in the media for human/animal in vitro fertilization (IVF) improves fertilization rate; addition of PPi in the semen extender for farm animal artificial insemination (AI) may improve pregnancy rates; furthermore, mammalian oocytes matured in vitro in a medium including PPi attain improved fertilization and developmental potential, while embryos cultured in medium supplemented with PPi have improved development to blastocyst.

The present invention provides a new apparatus/system for determining fertility of mammalian sperm based on a specific sperm-binding test that mimics sperm binding to an egg. The present invention relates to male infertility, and in particular to assays for predicting fertility in animals including human and bovines. In some embodiments, semen samples are evaluated by measuring the amount of ubiquitin in the sample, and in particular by measuring the extent of ubiquitination spermatozoa. Increased levels of ubiquitination in a sample are correlated with lower fertility. Ubiquitination may be assayed by several methods, including immunocytochemical measurement, ELISA, and flow cytometry.
