An isolated mutated green fluorescent protein (gfp) gene for chromosomal insertion into a bacterium, wherein the gene is capable of being expressed in bacteria and produce sufficient fluorescence under illumination from a UV lamp in a bacterial colony to be seen by the naked eye. The invention is directed to a method for obtaining a promoter from an organism using a proteomics approach, isolated promoters from fungal species and uses of the isolated promoters.

Effective management of finfish stocks to avoid or mitigate the threat of fish-killing phytoplankton is of increasing concern, particularly in temperate seas. Intensive spatial and temporal sampling is required to monitor and quantify potentially harmful species, so that prior warning can be received of an imminent bloom. The use of large-subunit rRNA (LSU rRNA)-targeted oligonucleotide probes based on the sandwich hybridization assay to detect the fragile species (Hada) Hada and Toriumi & Takano (Raphidophyceae) is disclosed. Species-specific sandwich hybridization assays were successfully developed for various Raphidophytes.
