Biocompatible coatings for medical devices are disclosed. Specifically, polymer coatings designed to control the release of bioactive agents from medical devices in vivo are disclosed wherein the solubility parameters of polymers and drugs are closely matched to control elute rate profiles. The present application also discloses providing vascular stents with controlled release coatings and related methods for making these coatings.

Biocompatible controlled release coatings for medical devices and related methods

The present disclosure generally relates to biocompatible polymers for coating or fabricating implantable medical devices and to implantable medical devices having the present biocompatible polymers. The disclosed biocompatible polymers exhibit superior biocompatibility and therefore minimize unwanted immune reaction from a patient into whom a medical device is implanted.

Disclosed herein are implantable medical devices comprising controlled release biodegradable block copolymers or coated with controlled release block copolymers and at least one drug releasable from the block copolymer. The controlled release block copolymers comprise least two blocks selected from the group consisting of polyesters, polyethers, and polyurethanes.

Biodegradable polymers useful for fabricating implantable medical devices and as coatings for medical devices are provided. The biodegradable polymers are biocompatible and can be tuned to provide optimum bioactive agent elution rates as well as degradation rates. The gradient coated stentof the present invention provides a coated stent having a continuous coatingdisposed on the stent elements. The present disclosure in a broad aspect provides for diazeniumdiolated phosphorylcholine polymers and associated methods for achieving nitric oxide release. The present polymers have superior biocompatibility and are useful for coating or fabricating medical devices such as a vascular stent.

The present invention provides a system for treating a vascular condition, including a catheter, a stent with a stent framework operably coupled to the catheter, and a drug-polymer coating on the stent framework including at least one plasticizer dispersed within the drug-polymer coating.

A drug delivery catheter and method for delivering microcapsules or microspheres containing a drug, pharmacological or other bioactive agent includes a balloon catheter having a coating on the balloon of a soluble biocompatible polymer containing a plurality of microcapsules or microspheres containing a drug or other agent. The balloon is porous to allow it to be inflated with a solvent fluid that passes through the porous wall of the balloon and dissolves the polymer, releasing the microspheres.

Specifically, methods for providing secondary amines-containing polymers using epoxide-opening reactions are provided. Disclosed are hindered amine nitric oxide (NO) donating polymers for coating implantable medical devices. The polymers include sterically hindered secondary amines that do not react with monomer carbonyls or electrophilic alkenes, facilitating the synthesis of the NO donating polymers. The polymers are coated on implantable medical devices, providing anti-restenosis therapy by the release of NO at the implantation site.

The methods of the present disclosure in a broad aspect provide for dry diazeniumdiolation procedures for producing nitric oxide releasing medical devices. These medical devices may alternatively have cap coats applied prior to dry diazeniumdiolation to produce nitric oxide releasing medical devices with cap coats.

N-substituted 4-aza-caprolactone biodegradable polymers, including derivatives thereof, useful for making implantable medical devices and coatings therefore are provided. The medical devices and coatings of the present invention can also be used for in situ controlled release drug delivery and are useful for treating or preventing medical conditions such as restenosis, aneurisms and vulnerable plaque.
