The invention generally concerns the use of amino acid denaturants for denaturing or separating double stranded nucleic acid molecules. More specifically, the present invention provides a method for the rapid isolation and recovery of a desired target DNA or RNA molecules from a mixture or library containing such molecules. The method involves the use of haptenylated probes and amino acid denaturants to select the desired molecules and eliminate the undesired library members from a sample. The invention also provides a method in which larger or full-length nucleic acid molecules can be isolated from the subpopulation of desired molecules.

Method for isolating and recovering target DNA or RNA molecules having a desired nucleotide sequence

An apparatus for washing magnetic particles including a well plate for receiving an array tube holder having x rows of tubes, each tube containing magnetic particles, the well plate having (x&plus;1) rows of wells for enabling the array tube holder to be shifted from a first position to a second position. The apparatus also includes ((x/2)&plus;1) rows of magnets asymmetrically positioned within the well plate adjacent to the rows of wells. Placing the array tube holder in the first position causes magnetic fields produced by each magnet to attract the magnetic particles in the x rows of tubes to one side of each tube, and shifting the array tube holder to the second position reverses the magnetic fields, causing the magnetic particles to stream across to the other side of each tube. The array tube holder may be a 96 tube array tube holder (8 rows by 12 columns) in which case, the well plate includes 108 wells (9 rows by 12 columns) for simultaneously washing 96 tubes.

Apparatus for washing magnetic particles

The present invention relates generally to methods for producing normalized nucleic acid libraries in which each member of the library can be isolated with approximately equivalent probability. In particular, the present methods comprise subtractive hybridization of a nucleic acid library with haptenylated (e.g., biotinylated, avidinated or streptavidinated) nucleic acid molecules that are complementary to one or more of the nucleic acid molecules of the library, such that the variation in the abundances of the individual nucleic acid molecules in the library is reduced. The invention also relates to production of normalized nucleic acid libraries (particularly cDNA libraries) in which contaminating nucleic acid molecules have been reduced or eliminated, and to normalized nucleic acid libraries produced by such methods.

Normalized nucleic acid libraries and methods of production thereof

An improved method is disclosed for diagnosing the presence of a chromosomal translocation characteristic of acute myelogenous leukemia. Nucleic acid molecules that may be used in this improved method are described.
