The present disclosure discusses a method of separating a sample (e.g., pharmaceutical drug, genotoxic impurity, biomarker, and/or biological metabolite) including coating a metallic flow path of a chromatographic system; injecting the sample into the chromatographic system; flowing the sample through the chromatographic system; separating the sample; and analyzing the separated sample using mass spectroscopy. In some examples, the coating applied to the surfaces defining the flow path is non-binding with respect to the sample—and the separated sample. Consequently, the sample does not bind to the low-binding surface of the coating of the flow path. The applied coating can increase the chromatographic peak area for the sample of the chromatographic system.

The present disclosure discusses a method of separating a sample (e.g., small organic acid metabolite) including coating a flow path of a chromatographic system. The quantified at least one peak of the extracted chromatogram and the quantified at least one band of the extracted spectrum are compared to a set of authentication criteria to determine authenticity of the botanical.

Methods for authenticating botanicals using a marker compound's related chromatographic profile and mass spectral profile jointly

A device for separating analytes is disclosed. The device has a sample injector, sample injection needle, sample reservoir container in communication with the sample injector, chromatography column downstream of the sample injector, and fluid conduits connecting the sample injector and the column. The interior surfaces of the fluid conduits, sample injector, sample reservoir container, and column form a flow path having wetted surfaces. A portion of the wetted surfaces of the flow path are coated with an alkylsilyl coating that is inert to at least one of the analytes. Use of vapor deposition coated flow paths for improved chromatography of metal interacting analytes

The present disclosure provides methods for the separation and quantitation of enantiomers of vitamin E using supercritical fluid chromatography (SFC) or carbon dioxide-based chromatography on chiral columns. The disclosed methods may be used to quantitatively determine the concentration of RRR-&#x3b1;-tocopherol in foods, food ingredients, dietary supplements, vitamin premixes, nutritional formulas, and medicines. A mixed-mode chromatography method for the determination of phosphorylated sugars in a sample is provided. The mixed-mode chromatography method includes obtaining a sample comprising at least one phosphorylated sugar. The sample is introduced onto a chromatography system. The chromatography system includes a column having a stationary phase material contained inside the column. The stationary phase material has a surface comprising a hydrophobic surface group and at least one ionizable modifier. The sample with a mobile phase eluent is flowed through the column, where the at least one phosphorylated sugar is substantially resolved and retained within seven minutes. The mobile phase eluent includes water with an additive and acetonitrile with the additive. The mobile phase eluent has a pH less than 6. The at least one phosphorylated sugar is detected using a detector.

Liquid chromatography/mass spectrometry methods for the analysis of polar molecules

An apparatus for use in a chromatography system includes a microfluidic substrate having a fluidic channel configured as an analytical chromatographic column and a fluidic port on one side of the microfluidic substrate. The fluidic port opens at a head end of the analytical chromatographic column. A dried blood spot (DBS) collection device holds one or more dried biological samples. The DBS collection device is directly coupled to the microfluidic substrate whereby one of the biological samples is placed into fluidic communication with the fluidic channel of the microfluidic substrate and an extraction of that biological sample flows toward the head end of the analytical chromatographic column. A diluent source fluidically coupled to the fluidic port supplies a solvent to the head end of the analytical column to dilute the extracted biological sample before the biological sample flows into the analytical chromatographic column.

Described are techniques for tuning parameters of a system. For one or more values of a distance, mass spectral analysis of a liquid analyte stream is performed. A corresponding ion signal intensity of a selected ion is obtained. The distance is measured from a first end of a first tube to a second end of a second tube which surrounds said first tube. Using a computing device, a current value of the distance is automatically adjusted in accordance with corresponding ion signal intensities obtained for the selected ion. Using a computing device, a desired value for the distance is automatically determined using the corresponding ion signal intensities. The desired value results in an ion signal intensity for the selected ion which is any of more than a threshold intensity and a maximum of ion signal intensities obtained by performing mass spectral analysis using different values for the distance.
