Screening assay for insecticides

A method and kit for detecting nucleic acid modification enzyme activity by monitoring the change in fluorescence polarization upon the change in mass of a fluorescently labeled oligo. A nucleic acid substrate comprises a template having a nucleic acid sequence annealed to a labeled oligo, wherein there is a nick or break located between the adjacent ends of the template and the labeled oligo. A nucleic acid modification enzyme is added to the substrate to form a reaction mixture. A stop reagent may or may not be used to further process the reaction mixture. A test compound is added to the reaction mixture, preferably prior to addition of the nucleic acid modification enzyme, whereby the resulting fluorescence polarization signal is compared to a standard signal to determine if the test compound inhibits nucleic acid modification enzyme activity.

Method and kit for identification for nucleic acid modification enzymes and inhibitors thereof

A parallel high throughput method and kit for the discovery of small molecule interactors of target proteins, in particular, target proteins expressed from entire genomes or subsets thereof. The target module is prepared by binding selectively a target protein, modified to allow its detection, to a surrogate ligand that is linked to an individually detectable bead. Sets of target modules are mixed together in each chamber of a multi-chamber container. A test compound or collection of test compounds is added to each chamber, and the interaction of a compound with each target module is observed. A compound that is specific for a particular target protein will displace that protein from its target module. The identity of the target module so disrupted is determined by identifying the particular bead to which the displaced target protein was attached.

Parallel high throughput method and kit

The invention relates to genes isolated from,that code for proteins essential for normal fungal growth and development. The invention also includes the methods of using these proteins to discover new fungicides, based on the essentiality of the gene for normal growth and development. The invention can also be used in a screening assay to identify inhibitors that are potential fungicides.
