The present invention relates to a murine homolog and a novel isoform of hD53, and a novel member of the D52 gene family, hD54. The genes and gene fragments of the present invention are themselves useful as DNA and RNA probes for gene mapping by in situ hybridization with chromosomes and for detecting gene expression in human tissues by Northern blot analysis.

Members of the D52 Gene family

The present invention relates to methods for identifying and selecting compositions useful in differentially modulating the expression of two or more mammalian genes, particularly matrix metalloproteinase (MMP) genes such as those encoding interstitial collagenase (and other genes comprising an AP1-binding site) and stromelysin-3 (and other genes comprising a retinoic acid response element (RARE)). In addition, the invention relates to methods of treating a mammal (such as a human) suffering from or predisposed to a physical disorder, using pharmaceutical compositions comprising the compositions identified or selected by the above-described methods. The methods of the present invention are useful in treating a variety of physical disorders in mammals including cancers (particularly carcinomas), inflammatory disorders, fibrotic disorders, ocular disorders and osteoporosis.

Methods and compositions for use in modulating expression of matrix metalloproteinase genes

The present invention relates to four novel human genes amplified and overexpressed in breast carcinoma and located on the q11-q21.3 region of chromosome 17. The four novel genes are useful in breast cancer prognosis. The present invention also relates to a fifth novel human gene expressed in breast carcinoma and located on chromosome 6q22-q23. A sixth novel gene is also described that is the murine homolog of the human D52 gene. The genes and gene fragments of the present invention are themselves useful as DNA and RNA probes for gene mapping by in situ hybridization with chromosomes and for detecting gene expression in human tissues (including breast and lymph node tissues) by Northern blot analysis.

The present invention relates to a gene encoding stromylsin-3, which is a new member of the metalloproteinase family. Expression of the stromelysin-3 gene has been found to be specifically associated with invasive breast, head, neck and skin cancer. The invention also relates to antibodies which specifically bind to human stromylsin-3 and the use of these stromelysin-3 antibodies for detection of the stromylsin-3 protein in a sample.

The present invention relates to a gene encoding a new member of the metalloproteinase family which has been found to be specifically associated with invasive breast cancer, and to methods of diagnosis for such cancer comprising detection of the marker or its nucleotide sequence, and to treatment or prophylaxis by inhibiting, altering the activity of or binding the marker, or by interfering with its synthesis.
