Embodiments of bridged tetrahydroisoquinolines and methods for their use in selectively inhibiting nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 2 are disclosed. The disclosed compounds have a structure according to general formula I or a pharmaceutically acceptable salt thereof: wherein '' represents a single or double bond, Ris hydrogen, halogen, lower aliphatic, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; Ris hydrogen, &#x2014;CHR, R, or &#x2014;SOR; Ris lower aliphatic, substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; Ris substituted or unsubstituted aryl, or substituted or unsubstituted heteroaryl; Ris lower aliphatic, or substituted or unsubstituted aryl; and Ris hydrogen, halogen, or lower aliphatic.

Tetrahydroisoquinolines as selective NADPH oxidase 2 inhibitors

The present invention relates to inhibitors of Nox1-dependent reactive oxygen species production and their use in the treatment of disorders associated with reactive oxygen species, such as hypertension and cancer.

Inhibitors of NOX1

A method is provided for treatment of a condition mediated by Nox2 in a patient. The method comprises administering to the patient by inhalation a polypeptide as described herein, able to inhibit superoxide production by Nox2. Conditions treatable in this manner include, without limitation, one or more of: right ventricular hypertrophy; pulmonary hypertension; acute lung injury; obstructive sleep apnea; ischemia/reperfusion injury in the lung; pulmonary fibrosis; an obstructive lung disorder such as Chronic Obstructive Pulmonary Disease (COPD), asthma, cystic fibrosis and emphysema; and atherosclerosis.
