A microfluidic device () comprises a substrate () having spatially defined and separated cell compartments () configured to accommodate cells. A respective first end () of the spatially defined and separated cell compartments () is in fluid connection with a flow input channel (). The microfluidic device () comprises at least one identification surface () comprising immobilized affinity molecules configured to capture cells of a species or serotype or of a group of species or serotypes.

Microfluidic device for cell characterization

A capturing of target cells from a biological sample is achieved by inducing a flow of the biological sample in a flow channel () of an upstream microfluidic device (). Target cells present in the biological sample are captured in cell channels () of the upstream microfluidic device (). Once at least a minimum number of target cells are captured in the cell channels (), the flow of the biological sample in the flow channel is reduced and are verse flow is applied at the upstream microfluidic device () to release the target cells captured in the cell channels () of the upstream microfluidic device () and enable transfer the target cells into cell channels () of a downstream microfluidic device ().

Cell capture in microfluidic devices

Phenotyping of cells involves loading a biological sample into a microfluidic device () comprising cell compartments () to capture biological material, including target cells, in the sample in the cell compartments (). A subset (A) of the cell compartments () is identified as comprising target cells exhibiting target phenotype characteristic(s) as determined based on monitoring biological material in the cell compartments () prior to addition of a test agent. The biological material is exposed to a test agent and a phenotypic response of the target cells to the test agent is determined based on monitoring target cells in the identified subset (A) of the cell compartments (). The phenotyping of the target cells is thereby not overshadowed by the response of other cells and non-cell material present in the biological sample.

Phenotypic characterization of cells

A microfluidic device () comprises a substrate () having a flow input channel () in fluid connection with a first fluid port () and a flow output channel () in fluid connection with a third fluid port () and cell channels () disposed between the flow input channel () and the flow output channel (). The cell channels () comprise a respective obstruction () designed to prevent the target cells from passing the respective obstruction () and into the flow output channel (). The microfluidic device () also comprises a pre-filter () with a filter channel () in fluid connection with a first filter port () and pre-filter channels () adapted to accommodate the target cells. A respective first end () of the pre-filter channels () is in fluid connection with the filter channel () and a respective second end () of the pre-filter channels () is in fluid connection with the flow input channel ().

Microfluidic device

This disclosure provides, among other things, a method for processing a membrane comprising rolling circle amplification (RCA) products. In some embodiments, this method may comprise: (a) obtaining a porous capillary membrane that comprises fluorescently labeled RCA products that are in or on the membrane; (b) depositing a curable polymer onto the membrane; and (c) curing the curable polymer to encapsulate the RCA products in a solid. In some embodiments, the curable polymer may be a silicone and may be transparent in its solid form. A kit for performing the method and a composition made by the method are also provided.

Method for processing rolling circle amplification products

A method of sample analysis is provided. In certain embodiments, the method may comprise: (a) filtering a liquid sample containing rolling circle amplification (RCA) products using a porous capillary membrane, thereby producing an array of the RCA products on the membrane; wherein the sample contains at least a first population of RCA products and a second population of RCA products, wherein the first and second populations of labeled RCA products are distinguishably labeled; and (b) determining the amount of the first labeled population of RCA products and the amount of the second labeled population of RCA products in an area of the membrane.

Use of a porous capillary membrane for determining the amount of rolling circle amplification products

A library of cell strains is characterized by culturing cells at spatially defined and separated positions in a culturing device () and determining a phenotypic characteristic of each cell strain in the culturing device (). The cells are fixated at the spatially defined and separated positions in the culturing device () followed by in situgenotyping a respective variable region () of each cell strain at the spatially defined and separated positions in the culturing device (). Each respective phenotypic characteristic is connected to each respective genotype based on the spatially defined and separated positions in the culturing device ().

Phenotypic characterization and in situ genotyping of a library of genetically different cells

The present invention is directed to a lateral flow assay device for the monitoring and measuring of coagulation and method thereof. Ideally, the invention is directed to a lateral capillary flow device for the monitoring and/or measurement of coagulation in a liquid sample wherein the device comprises a non-porous substrate with a zone for receiving a sample and a defined flow path zone wherein a clotting agent is deposited on at least part of the defined flow path zone to accelerate the coagulation of the liquid sample, enable the formation of an evenly distributed clot along the defined flow path zone and to result in the change in flow rate or cessation of flow of the liquid sample along the defined flow path zone.

Lateral flow assay device for coagulation monitoring and method thereof

A sensor comprising a substrate () having a first surface () and a second surface () is described. The first surface has at least one sensor site () provided thereon. The substrate is configured such that on excitation of a sample provided at the sensor site, luminescence originating from the sensor site propagates into the substrate, the second surface of the substrate being configured to selectively transmit the luminescence propagating within the substrates at angles greater than the critical angle out of the substrate where it may be detected by a detector () provided below the substrate.

Luminescence based sensor

A fluorescence reader for an optical assay arrangement that includes a polymeric sample substrate having a reaction site-surface and a substrate surface. The fluorescence reader includes a light source arranged to illuminate the reaction site-surface through the substrate surface, and a detector device arranged to detect fluorescent light emitted from the reaction site-surface and transmitted through the substrate surface, the substrate surface being configured to increase transmission of emitted fluorescent light by suppression of total internal reflection.

Fluorescence reader

A fluorescence reader for an optical assay arrangement includes a polymeric sample substrate having a reaction site-surface and a substrate surface. A light source is arranged to illuminate the reaction site-surface through the substrate surface, and a detector device is arranged to detect fluorescent light emitted from the reaction site-surface and transmitted through the substrate surface, the substrate surface provided, for example, as a light-collecting body to allow fluorescent light rays exceeding a critical emission angle for total internal reflection to escape the substrate and enter the body.

The invention relates to a micro fluidic system comprising a substrate, and, provided on said substrate, at least one flow path interconnecting with functional means in which liquid samples can be treated by desired procedures. The flow paths are laid out to form a pattern for the transport of liquid samples to and from said functional means. These flow paths comprise a plurality of micro posts protruding upwards from said substrate, the spacing between the micro posts being small enough to induce a capillary action in a liquid sample applied anywhere within any of said flow paths, so as to force said liquid to move from where said liquid sample was applied.

Micro fluidic structures

The invention comprises a polymeric microarray support () for an optical assay arrangement () comprising optical means () for detection of light emitted from the support. The microarray support is provided with microfeatures comprising a surface enlarging pattern (), i.e. grooves having a selected depth ().
