Provided are a system and a method for prediction of an intradialytic adverse event, where a machine learning model of two-class classification is utilized to predict intradialytic adverse events in quasi-real time, such that features extracted in an ongoing hemodialysis process in real time can have the hemodialysis session alerted for forthcoming adverse events. Therefore, clinicians can be warned to take necessary actions and adjust the hemodialysis machine settings in advance. In addition, a computer readable medium thereof is also provided.

The present invention provides a microfluidic bio-reactor device, which comprises: a first cell culture layer; a microfluidic base, which is located on the top of the first cell culture layer; a microfluidic layer, which is located on the top of the microfluidic base and have an air-bubble removal device; a microfluidic roof, which is located on the top of the microfluidic layer; the present invention also provides a method for culturing cells by the microfluidic bio-reactor device of the present invention and the kit with a cell-loading device and present microfluidic bio-reactor device.

Microfluidic bio-reactor device

The present invention describes an efficient and rapid protocol to generate hepatocyte cells from human induced pluripotent stem cells (iPSCs). According to the method, the iPS cells can be differentiated into functional hepatocyte cells in a short time course (less than 15-20 days). The iPSC-derived hepatocyte cells of the invention have a similar gene expression profile to mature hepatocytes. Moreover, the iPSC-derived hepatocyte cells is proved to rescue lethal fulminant hepatic failure in a non-obese diabetic severe combined immunodeficient mouse model. The rapid and efficient differentiation protocol for generation of iPSC-derived hepatocyte cells may offer an alternative option for treatment of liver diseases.

Methods for producing mature hepatocytes

The present invention provides a prism. The prism includes a lower surface, an upper surface, a first side surface and a second side surface. The first side surface and the second side surface are disposed between the upper surface and the lower surface. The first side surface and the second side surface of the prism are one-dimensional parabolic surfaces. The lower surface is used to receive light. The first side surface is used to reflect the light from the lower surface to the upper surface. The second side surface is used to reflect the light from the upper surface to the lower surface for further analysis in the process unit afterwards.

Coupling prism and optical detection system thereof

A method for preparing isolated hepatocytes is disclosed. The method comprises: a) culturing mesenchymal stem cells (MSCs) in a medium comprising hepatic growth factor (HGF) to cause the MSCs to differentiate toward hepatocytes, wherein the MSCs are isolated from bone marrow or umbilical cord blood; b) culturing cells from a) in a medium comprising HGF and oncostatin M (OSM) to facilitate the cell differentiation toward hepatocytes; and c) culturing cells from b) in a medium comprising OSM to cause the differentiated cells to mature into hepatocytes, and thereby producing the isolated hepatocyte cells.

Systems and methods for making hepatocytes from extrahepatic somatic stem cells and use thereof

A method for making hepatocytes from extrahepatic somatic stem cells comprises: a) culturing somatic stem cells in a medium comprising hepatic growth factor to cause the stem cells to differentiate toward hepatocytes; b) culturing cells from a) in a medium comprising HGF and oncostatin M to facilitate the cell differentiation toward hepatocytes; and c) culturing cells from b) in a medium comprising oncostatin M to cause the differentiated cells to mature into hepatocytes, thereby producing a cell population that has morphological features of hepatocytes and at least four of the following characteristics: i) antibody-detectable expression of albumin; ii) real-time reverse transcriptase-polymerase chain reaction-detectable expression of &#x3b1;-fetoprotein, HNF-1&#x3b1;, HNF-3&#x3b2;, HNF-4, HNF-6, &#x3b1;1-antitrypsin, alkaline phosphatase, tryptophan 2,3-dioxygenase, tyrosine aminotransferase, cytochrome P450 family 2 subfamily E polypeptide 1, glutamine synthetase, and/or low density lipoprotein receptor; iii) urea secretion; iv) cytochrome p450 enzyme activity; v) glycogen storage; and vi) low density lipoprotein uptake.
