Provided is a microscope system including: a plurality of objective lenses that are selectively used to collect fluorescence from a specimen; a detecting unit that obtains image information of the specimen on the basis of the fluorescence collected by the objective lens; and an adjustment unit that adjusts the observation range of the specimen S and that divides the observation range into one or more observation regions corresponding to a capture range captured by the detecting unit, wherein the adjustment unit calculates an observation magnification at which the entire well is included in the observation range, the number of observation regions is minimized, and the resolution becomes the highest, on the basis of information about the size of the well in a multi-plate for accommodating the specimen and information about an objective magnification of the objective lens, and adjusts the observation range on the basis of the calculated observation magnification.

It is possible to easily acquire an original image of a specimen that satisfies necessary conditions for generating a super-resolution image. The present invention provides a scanning microscope apparatus that includes an objective lens that irradiates a specimen with laser light emitted from a laser light source; a scanning portion that scans the laser light irradiated onto the specimen via the objective lens; and a PC that acquires an original image of the specimen, calculates, from the magnification of the objective lens, information about the number of pixels of the original image or information about the zoom magnification of the original image that achieves the per-pixel pixel resolution needed to generate a super-resolution image, and controls the scanning portion or image acquisition according to the calculated information about the number of pixels or information about the zoom magnification.

A scanning microscope includes a light source unit that projects laser light, an optical system that converges and applies the laser light onto data obtaining points on a specimen, and a data obtaining order deciding unit that decides a data obtaining order such that adjacent data obtaining points are not consecutive in the data obtaining order. A scanning unit scans the laser light in accordance with the data obtaining order, a detector detects detection light from the data obtaining points, respectively, and a storage unit stores luminance information on the detection light detected by the detector in association with positional information on the data obtaining points, respectively. An image formation unit forms a two-dimensional image based on the stored associated luminance and positional information.

An observation laser beam for observing a specimen and a manipulation laser beam for manipulating the specimen are multiplexed; the multiplexed beams irradiate the specimen, which is mounted on a stage, via an objective lens; and fluorescence emitted from inside the specimen in the observation optical axis direction is detected. In a preparation mode, a focal-position adjusting unit is controlled so that the focal position of the observation laser beam and the focal position of the manipulation laser beam are coincident, independent of the movement of the objective lens or the stage by a focusing mechanism; and in an observation mode, the focal-position adjusting unit is controlled so as to cancel out the shift of the focal position of the manipulation laser beam according to the movement of the objective lens or the stage.

A scanning laser microscope is configured to include: a scanner unit for performing two-dimensional scanning on a specimen using a light beam; a photodetector unit for detecting a light from the specimen; an A/D converter for converting an output signal from the photodetector unit into a digital signal; a CPU for controlling the scanner unit and generating image data of the specimen from the digital signal output from the A/D converter; a display unit for displaying the image data; an external device for outputting an external signal; and a data storage unit, connected to the external device, for recording the time when the external signal is output. With the configuration, the data stored in the data storage unit can be read by the CPU.
