Self-assembled gels and methods of making thereof are described. A selective range of ratios between an organic solvent and optionally water, or an aqueous solution, allows for combinations of gelators (such as GRAS amphiphiles or prodrug-based gelators), co-gelators (such as anti-retrovirals), and solvents to form self-supporting hydrogels or organogels. The resulting gels may be used in methods of treating HIV and/or hepatitis.

A self-assembled gel composition with enhanced adhesion to cartilage tissue is provided. A cationic agent co-self assembles with a generally regarded as safe (GRAS), low molecular weight (&#x3c;2,500 Da) gelator, forming homogeneous self-supporting gel that can encapsulate one or more therapeutic agents for controlled release. The composition adheres to connective tissue, e.g., cartilage, to a greater extent and a greater length of time than a self-assembled gel from gelators alone. The composition is used to specifically target connective tissue and deliver one or more therapeutic, prophylactic, or diagnostic agents for controlled release to improve dosing efficacy.

Self assembled gels for controlled delivery of encapsulated agents to cartilage

A non-covalently assembled hydrogel or organo-gel composition with serum stability is described. Low molecular weight (&#x3c;2,500 Da), generally regarded as safe (GRAS), materials assemble in the presence of a stabilizing agent at an appropriate molar percentage, forming hydrogel or organo-gel with nanostructures that resist disassembly or destabilization in serum for an extended period of time. The composition is used to deliver one or more therapeutic, prophylactic, or diagnostic agents, allowing for controlled release in response to biological stimuli such as enzymes and a greatly improved dosing efficacy.
