The present invention provides improved methods for maintaining the physical separation and identity integrity of a biological cellular sample from a patient during processing. The invention enables parallel processing of biological cellular samples, such as patient samples, in a space and time efficient fashion. The methods of the invention find particular utility in processing patient samples for use in cell therapy.

Cell processing method

In one aspect, the present invention relates to a methodfor identifying one or more phenotypes from a multi-parameter data set. The methodcomprises measuringcorrelation between pairs of parameters within the multi-parameter data set, modifyingcorrelated parameter values within a predetermined multi-parameter data analysis set to form an analysis parameter set, and analyzingthe multi-parameter data set using the analysis parameter set to identify one or more phenotypes from the multi-parameter data set. Various embodiments of the present invention may, for example, be used in an automated high-content screening (HCS) apparatusfor biological cellular analysis.

Method and apparatus for multi-parameter data analysis

Components are disclosed which include a first component section and a second component section joined to form a hollow structure defining a plenum having an interior surface, wherein the component sections each include mating ridges joined together along the length of the plenum, and a corrosion-resistant cladding layer including a corrosion-resistant material overlaying the interior surface of the plenum. In one embodiment, the component is a gas turbine combustor fuel manifold. A method of forming the components includes applying corrosion-resistant segments including a corrosion-resistant material to each of the surfaces of the component sections, and joining the component sections to form the component, wherein joining the component sections includes fusing the corrosion-resistant segments into the corrosion-resistant cladding layer, and joining the mating ridges of the component sections.

Component, gas turbine component and method of forming

The invention provides a system and method of analyzing the motion of a biological object, particularly the motion of cultured organisms or cell cultures. The system and method of the invention may be used to determine the effect of a physical stimulus or a test agent on the motion of cell cultures. The system and method is of particular use in assessing the effect a chemical may have on the contractile motion of cardiomyocyte cell cultures.

System and method for determining motion of a biological object

Apparatus and methods are provided for performing cell growth and cell based assays in a liquid medium. The apparatus comprises a base plate supporting a plurality of micro-channel elements, each micro-channel element comprising a cell growth chamber, an inlet channel and an outlet channel, a cover plate positioned over the base plate to define the chambers and connecting channels. Means are incorporated in the cell growth chambers, for cell attachment and cell growth. In particular, the invention provides a rotatable disc microfabricated for performing cell growth and cell based assays. The apparatus and method can be used for the growth of cells and the detection and measurement of variety of biochemical processes and products using non-invasive techniques, that is techniques which do not compromise the integrity or viability of cells.

Microfabricated apparatus for cell based assays

A method of analysing a plurality of biological entities using an imaging apparatus. The method comprises: providing a marker for said plurality of biological entities, said marker being capable of identifying objects within said plurality of biological entities when detected using the imaging apparatus, the method of provision being arranged such that said marker is capable of identifying said objects during a first time period, and said marker is less capable of identifying said objects during a second time period; during the first time period, recording a marked-up image in which spatial definitions of said objects are identifiable from said marker; during the second time period, recording a first image of said plurality of biological entities; and generating a spatial definition for an object in said first image using data derived from said marked-up image.

The present invention relates to polypeptide and nucleic acids constructs which are useful for determining the cell cycle status of a mammalian cell. Host cells transfected with these nucleic acid constructs can be used to determine the effects that test agents have upon the mammalian cell cycle.

Cell cycle phase markers

The present invention provides a cell imaging technique for automatically tracking the progression of a cell through the cell cycle over time through segmentation of a volume of two-dimensional time-lapse images. The technique allows long-term tracking of the cell cycle progression of an individual cell or multiple cells. Further, the invention provides a unique display of cell cycle progression, allowing an end user to easily determine changes to cell cycle progression for a cell of interest.

The present invention relates to non-destructive and dynamic means for determining the cell cycle position of living cells. The invention provides stable cell lines which can be used to determine the cell cycle position, together with methods for measuring the effect of a test agent on the cell cycle position.

Disclosed is a method for increasing the fluorescence of a Cyanine dye molecule comprising at least one NOgroup characterized by the reduction of the at least one NOgroup to NHOH or NHby the action of a nitroreductase. The cyanine dye molecule comprising at least one NOgroup can be used as a substrate for detecting nitroreductase enzyme activity in a composition and allows for the use of a nitroreductase enzyme in an enzyme-reporter system for the detection of analytes, binding reactions or gene expression.

Fluorescent detection method and reagent

Disclosed is method for measuring a change in a cellular morphological parameter, for example, the position and/or the shape of a cell. The method comprises labelling a population of cells with a marker substance which is in a non-detectable state, but which may be caused to become detectable by exposure to light of a suitable first wavelength. By exposing a sub-population of labelled cells to light of an appropriate first wavelength in a predetermined pattern, the sub-population of cells may be defined in which the area covered by the sub-population conforms to the pattern of illumination used. Subsequent examination of at least a portion of the population of cells using illuminating light of a second wavelength selected to be suitable for detection of the marker, will reveal patterns of distribution of the marker substance indicative of a change in said cellular morphological parameter. Also described and claimed is a method for screening for a test agent whose effect on the cellular morphological parameter is to be determined.

Method for the determination of changes in a cellular morphological parameter

The present invention provides novel engineered derivatives of green fluorescent protein (GFP) which have an amino acid sequence which is modified by amino acid substitution compared with the amino acid sequence of wild type Green Fluorescent Protein (SEQ ID NO: 2). The modified GFPs exhibit enhanced fluorescence relative to wtGFP when expressed in non-homologous cells at temperatures above 30&#xb0; C., and when excited at about 490 nm compared to the parent proteins, i.e. wtGFP. An example of a preferred protein is F64L-S175G-E222G-GFP. The modified GFPs provide a means for detecting GFP reporters in mammalian cells at lower levels of expression and/or increased sensitivity relative to wtGFP. This greatly improves the usefulness of fluorescent proteins in studying cellular functions in living cells.

Fluorescent proteins and methods of using same

The invention provides a novel, non-destructive and dynamic process for determining the cell cycle position of living cells. The invention also provides DNA constructs, and cell lines containing such constructs, that exhibit activation and deactivation of a detectable reporter molecule in a cell cycle specific manner. The invention thus allows greater precision in determining cell cycle phase status than existing techniques and further provides a method for continuous monitoring of cell cycle progression in individual cells.
