Detection method and apparatus for detecting microbial growth

Embodiments of the present invention provide an apparatus and a method for separating particles within a sample. The apparatus includes a reservoir for receiving a biological specimen therein, wherein the specimen includes a plurality of different sized particles in a liquid. At least one physical state of the specimen is capable of being controlled in order to initially separate at least a first type of particles under the influence of gravity. The apparatus also includes a rotatable disk positioned proximate to the reservoir and configured to receive the first type of particles thereon. The disk has holes defined therethrough that are configured to further separate the first type of particles based on particle size. The apparatus includes a mechanism configured to rotate the disk, and at least one collection tube positioned proximate to the disk and configured to receive at least a portion of the first type of particles passing through respective holes defined in the disk and/or around the disk in response to rotation of the disk.

Apparatus and method for separating particles within a specimen

The early prediction or diagnosis of sepsis advantageously allows for clinical intervention before the disease rapidly progresses beyond initial stages to the more severe stages, such as severe sepsis or septic shock, which are associated with high mortality. Early prediction or diagnosis is accomplished using a molecular diagnostics approach, involving comparing an individual's profile of biomarker expression to profiles obtained from one or more control, or reference, populations, which may include a population who develops sepsis. Recognition of features in the individual's biomarker profile that are characteristic of the onset of sepsis allows a clinician to diagnose the onset of sepsis from a bodily fluid isolated at the individual at a single point in time. The necessity of monitoring the patient over a period of time is, therefore, avoided, advantageously allowing clinical intervention before the onset of serious symptoms. Further, because the biomarker expression is assayed for its profile, identification of the particular biomarkers is unnecessary. The comparison of an individual's biomarker profile to biomarker profiles of appropriate reference populations likewise can be used to diagnose SIRS in the individual.

Diagnosis of sepsis or SIRS using biomarker profiles

Mass spectrometry techniques for determining the status of sepsis in an individual are provided. A biomarker profile resolved from a biological sample, taken from the individual, using a mass spectrometry technique is compared to a reference biomarker profile. A single such comparison classifies the individual as belonging to or not belonging to a reference population. The individual's biomarker profile and the reference biomarker profile comprise a plurality of ions each having a mass-to-charge ratio of about 100 Daltons to about 1000 Daltons. The plurality of ions can be detected by electrospray ionization mass spectrometry in positive mode. The comparison uses a decision rule, such as a classification tree, that determines the status of sepsis in the individual without requiring knowledge of the identity of the biomarkers in the biomarker profile from the individual and without requiring knowledge of the identity of the biomarkers in the reference biomarker profile.

Method of predicting the onset of sepsis in SIRS-positive individuals using mass spectrometry

A test strip reader is provided with an optical mouse integrated circuit sensor that reads the test line, reference line and control line of a lateral flow assay strip by light absorption and determines the position of the test strip beneath the image on the integrated circuit board using digital signal processing algorithms. The digital signal processing algorithms process the imaged surface at a very high rate (e.g., 1500 frames per second) to determine the direction and speed of movement of the lateral strip with respect to the reader optics and image capture components. The sensor outputs this position information in a quadrature pattern format. A microcontroller uses the known position of the strip with respect to the reader to calculate the optical absorption of each colored reagent line, and the diagnostic significance (i.e., either qualitative or quantitative) of the test strip.

Apparatus and method for using optical mouse engine to determine speed, direction, position of scanned device and to obtain quantitative or qualitative data from same

A system and method employing infrared laser spectrography and dual wavelength modulation to monitor the concentration of a gas, such as oxygen or carbon dioxide, in the sample vial, or to monitor the pressure in the sample vial, to thus detect for microorganism growth in the sample vial. The system and method each employ an energy emitting device, such as an infrared laser, a detector and a signal analyzer, such as a spectroscopy device. The infrared laser emits toward the container infrared energy having a substantially single wavelength substantially equal to a wavelength at which the gas absorbs the infrared energy. The detector detects a portion of the energy signal that passes through the container, and the signal analyzer spectroscopically analyzes the detected portion of the energy signal to determine whether the gas exists in the container, or to determine the pressure in the container. The system and method thus determines whether an organism or cell of interest is present or viable in the container based on the results of the gas or pressure detection.

System and method for optically monitoring the concentration of a gas, or the pressure, in a sample vial to detect sample growth

A system and method for monitoring the concentration of a medium in at least one container using photothermal spectroscopy. The medium can be a gas, such as oxygen or carbon dioxide, or a solid or liquid. The system and method each employs an energy emitting device, such as a laser or any other suitable type of light emitting device, which is adapted to emit a first energy signal toward a location in the container. The first energy signal has a wavelength that is substantially equal to a wavelength at which the medium absorbs the first energy signal so that absorption of the first energy signal changes a refractive index of a portion of the medium. The system and method each also employs a second energy emitting device, adapted to emit a second energy signal toward the portion of the medium while the refractive index of the portion is changed by the first energy signal, and a detector, adapted to detect a portion of the second energy signal that passes through the portion of the medium. The system and method each further employs a signal analyzer, adapted to analyze the detected portion of the second energy signal to determine an amount of a sample in the container based on a concentration of the medium in the container. In particular, the signal analyzer can analyze the detection portion of the second energy signal to determine whether the sample includes an organism which consumes or emits the medium.
