Delivery of small interfering RNA and micro RNA through nanogels containing hydrophobic pseudo-peptides

In some aspects, methacrylate co-polymers crosslinked with an enzymatically cleavable peptide linker are provided and may be used for the oral delivery of a therapeutic. The peptide linker may be cleavable by an enzyme in the small intestine and may allow for the delivery of a therapeutic protein or nucleic acid to the small intestine. Also provided are methods of using the polymers for the treatment of a disease.

Hydrogels for delivery of therapeutic compounds

In some aspects, improved hydrogel copolymers (e.g., comprising itaconic acid and N-vinylpyrrolidone) are provided and may be used, e.g., for oral delivery of a therapeutic protein. In some embodiments, improved methods for loading a therapeutic protein (e.g., a high isoelectric point protein) into a hydrogel copolymer are provided, and may comprise incubating the therapeutic protein and the hydrogel in a reduced ionic strength loading solution. In some embodiments, use of the reduced ionic strength loading solution can result in hydrogel copolymer-therapeutic protein compositions that display improved pharmacokinetic attributes, e.g., improved loading and/or release of a therapeutic protein.

Polymers for delivery of therapeutic proteins

In some aspects, a composition comprising a pH-sensitive crosslinked copolymer of methacrylic acid and poly(ethylene glycol) monomethyl ether methacrylate and a therapeutic protein is provided. In some embodiments, the therapeutic protein is a high molecular weight protein such as factor VIII or factor IX. In some embodiments, the composition is orally administered to a patient to treat a disease or disorder such as, e.g., hemophilia.

Such sensors may comprise the recognitive hydrogels and an impedance sensing component. Such methods may comprise providing a triggering molecule, providing a sensor comprising a molecularly imprinted polymer having a binding cavity specific for the triggering molecule, a conductive polymer associated with the molecularly imprinted polymer, and an impedance sensing component, introducing the triggering molecule into the sensor, and detecting a change in impedance of the recognitive hydrogel with the impedance sensing component.

A detector is operatively connected to the microcantilevers for sensing a state of deformation thereof. On each microcantilevers is deposited a layer of an environmentally sensitive hydrogel polymer having a configuration changing in accordance with presence of an environmental parameter.
