The invention is directed to a suppository based vaccine delivery system for immunizing against urogenital and anorectally transmitted infectious disease in humans and animals and a method for treating the same. More particularly, this invention is directed to a suppository based vaccine delivery system for the prophylaxis against or treatment of urogenital or anorectal transmitted infectious diseases, such as from viral or microbial pathogens. The suppository based delivery system comprises vaccine and/or vaccine adjuvant(s) comprised of whole or fractionated viral or other microbial pathogens, or their purified cellular constituents, whether native, mutated, synthetic, cloned or recombinantly expressed, that consists of nucleic acids, proteins, lipids or other antigenic determinants capable of producing humoral or cellular-mediated immunity in humans or animals; and a polyethylene glycol base; wherein the suppository is adapted to be inserted into a bodily orifice of a human or animal so as to allow the suppository to be in contact with tissue of the bodily orifice to facilitate transfer of vaccine or vaccine adjuvant(s) material therethrough.

Urogenital or anorectal transmucosal vaccine delivery system

This invention is directed to a suppository-based vaccine delivery system for immunizing against urogenital infectious disease in humans and a method for treating same. More particularly, this invention is directed to a suppository-based vaccine delivery system for the prophylaxis against urogenital infectious diseases, such as urinary tract infections. The suppository-based vaccine delivery system is comprised of a vaccine comprised of inactivated bacteria which originate from cultures of 8 to 14 uropathogenic bacteria strains of the species: Escherichia coli, Klebsiella pnieumoniae, Proteus mirabilis, Proteus morganii, and Streptococcus faecalis; and polyethylene glycol suppository base; wherein the suppository is adapted to be inserted into a bodily orifice of a human so as to allow the suppository to be in contact with tissue of the bodily orifice to facilitate transfer of suppository material therethrough.

Vaginal suppository vaccine for urogenital infections

By reacting 1-5.5M nitric acid with from 45 to 170 mmol of primary C.sub.1 -C.sub.5 -alkanol per l of nitric acid, an improved preparation is obtained for the local treatment of changes in skin and mucous membrane and of infections of the skin and mucous membrane. The preparation comprises 1-5.5M nitric acid, not more than 170 mmol of C.sub.1 -C.sub.5 -alkanoic acid per l of solution, and nitrate reduction products in a concentration corresponding to from 0.1 to 6 mg of nitrite per ml of solution. The preparation is notable for a good activity and a low risk of side effects.

Preparation for skin and mucous membrane

A method of introducing effectively non-anionic agents into mammalian red blood cells without unacceptable loss of cell contents, utilizing an osmotic pulse technique wherein an aqueous diluent medium is provided containing the desired effectively non-anionic agent(s) and an effective amount of water-soluble polyanion. The polyanion is non-disruptive to the lipid outer membranes of red blood cells and contains at least two anionic moieties per molecule. The method comprises suspending packed red blood cells in an aqueous solution containing a compound which readily diffuses into and out of the cells, rapidly creating a trans-membrane osmotic gradient by diluting the solution containing the cells in suspension with an essentially isotonic aqueous medium containing an effective amount of polyanion and one or more effectively non-anionic agents to be introduced, thereby causing diffusion of water into the cells with consequent swelling and increase in permeability of the outer membranes, and maintaining the increase in permeability for a period of time sufficient only to permit transport of the desired agent and polyanion into the cells and diffusion of the compound out of the cells. In another embodiment, for use where a polyanion in the aqueous diluent medium would be undesirable, the red blood cells are subjected to shear stress immediately after the osmotic pulse occurs, resulting in resealing of the outer membranes of the cells and regaining their original size and shape, with the desired agent(s) incorporated therein.

Method for introducing desired agents into red blood cells

A process and medium are disclosed for the lyophilization of red blood cells which comprises the use of solutions including monosaccharide hexoses and pentoses, biocompatible polymers and polyanions to permit the reconstitution of viable red blood cells.

Lyophilization of red blood cells

The composition consists of an aqueous solution of nitric acid and nitrous acid with a pH below 1 and an acid equivalent of from 6 to 10 mmole/ml; the amount of nitrous acid corresponds to 0.01 to 5 mg of nitrite (NO.sub.2.sup.-) per ml. Additionally, the solution preferably contains acetic acid, oxalic acid and lactic acid as well as copper and cadmium ions. For the preferred composition, the preparation takes place by mixing the components and letting them react to the end or by an oxidative method, in the course of which nitrous acid is formed. The composition is suitable for the local treatment of superficial lesions of the skin and mucous membranes in the fields of cosmetics and medicine, particularly for certain malignant tumors. Experimentally, the effectiveness can be checked by two biological tests developed for this purpose.

Use of dilute nitric acid solutions for treating certain skin lesions

Method and apparatus for creating reversible intracellular hypertonicity in red blood cells of mammals in order to introduce desired materials into the cells or achieve therapeutically desirable changes in the characteristics of intracellular hemoglobin. A packed red blood cell fraction is incubated in a solution containing a compound which readily diffuses into and out of the cells; a trans-membrane osmotic gradient is rapidly created by contacting the cells with a near-isotonic aqueous medium with or without a desired agent to be introduced, causing diffusion of water into the cells with consequent swelling thereof and permitting diffusion of said compound out of the cells; and the cells are then separated and washed to obtain a sterile fraction of substantially intact cells.

Method for altering red blood cell function and survival

Lipid vesicles, such as those containing inositol hexaphosphate useful for improving the oxygen-release of intact erythrocytes, may be rendered stable to long term storage at temperatures below the freezing point of the encapsulated solution, by rapid freezing at a temperature of from about -196.degree. C. to about -30.degree. C.
