Methods and compositions for preparing and priming a tissue graft for an accelerated therapeutic effect are provided herein. In one embodiment, the method includes obtaining a tissue containing viable cells from a donor, wherein the viable cells are endogenous to the tissue and remain resident in the tissue; and priming the viable cells with one or more stimuli comprising simulated hypoxia to produce a primed tissue, wherein when grafted to a recipient the primed tissue provides a benefit compared to non-primed tissue.

Methods and compositions for preparing transplant tissue

Methods and compositions for preparing and priming a tissue graft for an accelerated therapeutic effect are provided herein. The invention is directed toward an improved formable bone composition for application to a bone defect site to promote new bone growth at the site which comprises a new bone growth inducing compound of demineralized lyophilized allograft bone particles ranging from about 100 to 850 microns. The bone particles are mixed in an excipient carrier combination containing carboxymethylcellulose, sodium hyaluronate, and a sodium phosphate saline buffer, the carboxymethylcellulose component of the carrier ranging from about 5.0 to about 11.0% of the composition and the sodium hyaluronate component of the carrier ranging from about 0.3 to about 0.7% of the composition, the composition having a pH between 6.5-7.5.

Method kit for treating bony defects

A construct for repairing articular cartilage defects includes a cap member and a base member. The cap member has an upper section and a stem depending from a central region thereof. The upper section includes a peripheral region. The stem includes a cavity. The base member has first and second ends, the first end including an annular recess dimensioned such that the stem is receivable therein. The first end also includes an annular edge positioned laterally outwardly from the annular recess, for abutting and supporting the peripheral region of the upper section when the stem is received in the annular recess. The base member further includes an island which is surrounded by the annular recess and receivable in the cavity of the stem, such that the island is abuttable with and may thereby support the central region of the upper section when the stem is received in the annular recess.

An implantable member for use in the body is provided herein. This implantable member includes a porous biocompatible substrate; the substrate having at least one surface sealed fluid-tight with self-aggregating protein particles of substantially the same diameter range. The self-aggregated protein particles are formed from a deposited aqueous slurry of the protein particles.

The invention is directed toward a cartilage repair assembly comprising a shaped allograft structure of subchondral bone with an integral overlying cartilage cap which is treated to remove cellular debris and proteoglycans and milled allograft cartilage in a bioabsorbable carrier. The shaped structure is dimensioned to fit in a drilled bore in a cartilage defect area so that either the shaped bone or the cartilage cap engage the side wall of the drilled bore in an interference fit and is in contact with a milled cartilage and biocompatible carrier mixture allowing cell transfer throughout the defect area. A method for inserting the shaped allograft structure into a cartilage defect area is also disclosed.

The invention is directed toward a sterile cartilage defect implant material comprising milled lyophilized allograft cartilage pieces ranging from 0.01 mm to 1.0 mm in size in a bioabsorbable carrier taken from a group consisting of sodium hyaluronate, hyaluronic acid and its derivatives, gelatin, collagen, chitosan, alginate, buffered PBS, Dextran or polymers with allogenic chondrocytes or bone marrow cells in an amount exceeding the natural occurrence of same in hyaline cartilage and adding a cell growth additive.

The invention is directed toward a cartilage repair assembly comprising a cylindrically shaped allograft structure of subchondral bone with an integral overlying smaller diameter cartilage cap which is treated to remove cellular debris and proteoglycans. The shaped structure is dimensioned to fit in a drilled bore in a cartilage defect area so that the subchondral bone of the structure engages the side wall of the bone portion of the drilled bore in an interference fit while the cartilage cap is spaced from cartilage portion of the side wall of the drilled bore forming a gap in which a milled cartilage and biocompatible carrier mixture is placed allowing cell transfer throughout the defect area. A method for inserting the shaped allograft structure into a cartilage defect area is also disclosed.

There is described a bio-compatible aqueous slurry for forming a fluid-tight barrier on a surface of a porous implantable article. This aqueous slurry includes a self-aggregating protein comminutate having a substantially uniform particle size, a bio-compatible plasticizer and water. This aqueous slurry is further characterized by having a viscosity of about 8,000 centipoise to about 60,000 centipoise at 25&#xb0; C. and a pH sufficient to maintain the protein suspended in the slurry.

The invention is directed toward a malleable bone putty and a flowable gel composition for application to a bone defect site to promote new bone growth at the site which comprises a new bone growth inducing compound of demineralized lyophilized allograft bone powder. The bone powder has a particle size ranging from about 100 to about 850 microns and is mixed in a high molecular weight hydrogel carrier, the hydrogel component of the carrier ranging from about 0.3 to 3.0% of the composition and having a molecular weight of about at least 10,000 Daltons. The composition contains about 25% to about 40% bone powder and can be additionally provided with BMP's and a sodium phosphate buffer.
