An electrophoresis medium membrane comprises a planar support, a planar cover sheet, and a layer of an electrophoresis gel medium provided between the planar support and the planar cover sheet. The electrophoresis gel medium contains an aqueous polyacrylamide gel, which is prepared by cross-linking polymerization of an acrylamide compound and a cross-linking agent in the presence of water and a compound having at least one carbamoyl group as a denaturing agent (or modifier). The layer of the electrophoresis gel medium has both a predetermined gradual change (gradient) in layer thickness and a predetermined gradual change (gradient) in concentrations of the acrylamide compound and the cross-linking agent.

Electrophoresis medium membrane

A means for electrophoresis suitably employable for analysis of biopolymers such as proteins, as well as for determination of base sequence of DNA, RNA, their fragments, and their derivatives is disclosed. The means comprises a membrane of an aqueous polyacrylamide gel for electrophoresis which is sandwiched between a plane support and a plane covering sheet. The membrane has a thickness gradient in its electrophoresis direction.

Means for electrophoresis

A method of making an electrophoresis medium having a predetermined gradient in concentrations of an acrylamide compound and a cross-linking agent comprises the steps of mixing two kinds of aqueous solutions containing the acrylamide compound and the cross-linking agent in different concentrations such that a mixing ratio of the aqueous solutions to each other is changed gradually, thereby to form a predetermined gradual change (gradient) in concentrations of the acrylamide compound and the cross-linking agent in the mixture, and carrying out cross-linking polymerization of the mixture in the presence of a polymerization initiator. Viscosity values of the two kinds of the aqueous solutions containing the acrylamide compound and the cross-linking agent in different concentrations prior to the mixing are adjusted to be substantially equal to each other.

Method of making an electrophoresis medium

In a medium for electrophoresis suitably employable for determination of base sequence of DNA, RNA, their fragments, and their derivatives, which comprises an aqueous polyacrylamide gel formed by crosslinking polymerization of an acrylamide compound and a crosslinking agent in the presence of water and at least one compound having a carbamoyl group, the improvement wherein glycerol is contained in the medium in a small amount ranging from 0.1 to 1.0 wt/v %.
