Provided are a fusion protein and a construction method thereof. The fusion method consists of: aprotein D and a Hin47 (Htra) protein. The fusion protein can serve as a protein vehicle for apolysaccharide-protein conjugate vaccine, thereby increasing immunogenicity of a polysaccharide antigen.

The invention provides an immunogenic composition for preventing pneumococcal diseases, comprising PspA-MRX1, PspA-EF5668, PspA-EF3296 and PlyL460D, wherein the amount of each component is 10-100 &#x3bc;g/ml; the immunogenic composition is prepared by adding to aluminum adjuvant the corresponding dosages of the four stock solutions PspA-mRX1, PspA-EF5668, PspA-EF3296 and PlyL460D, and mixing them homogeneously; the immunogenic composition can prevent infection and invasion by, covering more than 95% of the strains in clinic.
