An individualized bacterial treatment of cancer is provided. The treatment includes a strain of bacteria modified by in-vivo passage through tumor grafts in experimental animals, where the modified strain exhibits enhanced cancer cell-targeting of a specific malignancy arising in a unique individual to the corresponding parent strain of bacteria. The treatment uses this modified strain for the treatment human solid-tumor malignancies by inoculating an individual with a quantity of the strain; and repeating inoculations at periodic intervals where repeated inoculations tend to progressively eliminate the solid tumor malignancy in the individual.

Individualized bacterial treatment of pancreatic cancer

Bacteria that are auxotrophic for at least two amino acids found in at least one tumor are effective anti tumor treatments, labeling agents, and vaccines against infection. Improved antitumor effects can also be provided such strains by passage through an appropriate tumor model.

Cancer selective auxotrophs

Methods to obtain genetic modifications of cells in histoculture are described. Modification is assisted by treating the histoculture with collagenase prior to contacting the histoculture with the delivery vehicle for the desired gene. Hair follicles and other organized tissues can be modified in this was and then transplanted into intact recipients.

Methods for introducing genes into mammalian subjects

A composition and method for treating disorders related to tyrosinase gene expression and melanin biosynthesis is disclosed. The composition comprises a tyrosinase encoding nucleotide sequence and an ORF-438 encoding nucleotide sequence derived from Streptomyces, adapted for expression in mammalian cells. Also disclosed is a model system for evaluating agents that affect pigmentation.
