A solid-phase permethylation procedure is described. For example, solid-phase permethylation can be utilized to prepare permethylated linear and branched, neutral and sialylated oligosaccharides, which can be analyzed by MALDI-MS.

A device for placement of effluent comprises a substrate positioner, a deposition conduit, and a conduit positioner. The substrate positioner supports and positions a substrate on which effluent exiting from the deposition conduit is to be deposited. A fluid treatment device is disclosed for mixing first and second fluids. The first fluid may be a treatment liquid that is contained in a container and that is used to change a property, such as the pH, density or ionic strength, of the second fluid. In some illustrative embodiments, the fluid treatment device is used to create an effluent by adding a buffer material and/or a matrix material to an analyte for subsequent MALDI MS analysis of the effluent. A mixing space is defined in a container between ends of first and second conduits. In some embodiments, a junction member couples together the ends of the first and second conduits. The junction member has a main passage that receives the ends of the first and second conduits. The junction member has at least one opening that provides fluid communication between an interior region of the container and the mixing space.

Polar, monolithic CEC columns having an amine and/or cyano functional stationary phase are used to separate complex mixtures of saccharides. Using the positive-ion mode of detection for neutral saccharides, detection conditions were optimized to low-femtomole sensitivities with the use of an ion trap mass spectrometer. The described column technology provides a universal system for separating a wide range of carbohydrates: mono- and oligosaccharides with the intact reducing end, as well as saccharide alditols.

Compounds containing .alpha.-discarbonyl groups are rendered capable of fluorescent emission by conjugation with carboxylic acid-terminated ortho-diaminoaryl reagents. Surfaces of silica-containing materials, such as the inner walls of silica capillaries, used in chromatographic, particularly electrophoretic, separations are coated with an organic polymer layer to reduce or eliminate surface charges. The layer is applied by first converting the silanol groups on the surface to silicon halide groups, then reacting these groups with an organometallic reagent having a terminal ethenyl moiety, preferably vinyl or allyl lithium or a vinyl or allyl magnesium halide, to convert the silicon halide groups to Si--R groups where the R retains the terminal ethenyl moiety, and finally reacting these ethenyl groups newly attached to the surface with a neutral organic monomer in an addition polymerization reaction to form a monomolecular noncrosslinked polymer layer over the surface. The resulting polymer layer is linked to the silica directly through a Si--C bond which is stable over a wide range of pH conditions.

Gels are formed in capillary tubes by electrophoretically drawing a charged polymerization initiator into a capillary tube pre-filled with monomer, crosslinking agent and catalyst. Selective monomolecular layers on siliceous surfaces used for the construction of high-resolution chromatographic columns are prepared by surface reactions with monohalogenated silanes, silazanes or silylamines, or monoalkoxy- or monoacetoxysilanes, and subsequent chemical modifications. Such monolayers form a basis for efficient films of chemically compatible stationary phases by introducing selective groups in gas-liquid chromatography and all forms of liquid chromatography, or for adsorbent modifications in gas-solid chromatography.
