An intravitreal injection system for administering a pharmacological agent formulation to an intravitreal compartment of an eye, comprising (i) an injection member coupled to, or comprising, an internal formulation chamber that is adapted to receive and contain the pharmacological agent formulation therein, (ii) a needle having a first end that is in communication with a formulation chamber and a second injection end, and (iii) a movable platform for positioning the system on the eye, guiding the needle, and limiting the penetration depth of the needle into the eye.

Intravitreal injection device and system

A transdermal electrotransport drug delivery device having an anode, a cathode and a source of electrical power electrically connected to the anode and the cathode. At least one of the anode and the cathode includes an electrode and a reservoir comprised of a housing composed of a polymeric material and an aqueous medium in contact with the housing. The aqueous medium includes (i) a drug or an electrolyte salt or a mixture thereof, (ii) propylene glycol, and (iii) an antimicrobial agent in an amount sufficient to inhibit microbial growth in the aqueous medium. The propylene glycol prevents the antimicrobial agent from being adsorbed by other materials used in the construction of the delivery device. A process for preparing a transdermal electrotransport drug delivery device is also provided.

Transdermal electrotransport delivery device including an antimicrobial compatible reservoir composition

This invention relates to a method for inhibiting a decrease in the transdermal flux of an agent that is being transdermally delivered or sampled over a prolonged period of time wherein the delivery or sampling involves disrupting at least the stratum corneum layer of the skin to form pathways through which the agent passes. The desired result is achieved by co-delivering or co-sampling the agent with an amount of at least one anti-healing agent wherein the amount of the anti-healing agent is effective in inhibiting a decrease in the agent transdermal flux compared to when the delivery or sampling of the agent is done under substantially identical conditions except in the absence of the anti-healing agent(s).

Methods for inhibiting decrease in transdermal drug flux by inhibition of pathway closure

An electrotransport composition comprises at least one C.sub.2 -C.sub.4 lower alcohol, unsaturated derivatives thereof, or mixtures thereof, and at least one C.sub.8 -C.sub.4 higher alcohol, unsaturated derivatives thereof, or mixtures thereof. The invention is directed to a composition and product for use in an electrotransport device. The composition includes an ionized drug, an anti-inflammatory agent and a solvent. Preferably, the anti-inflammatory agent is hydrocortisone or esters thereof, and the solvent is an alcohol, a glycol, a glycerol, a cyclodextrin, a nonionic surfactant or mixtures thereof. The composition preferably further includes a gel matrix that is a swellable, hydrophilic polymer, a water soluble hydrophilic polymer or a solid polymer. A hydrophobic polymer and/or additives may also be included.

An electrotransport drug delivery device which delivers an anti-inflammatory agent in order to reduce skin irritation associated with electrotransport delivery of the drug is provided. The anti-inflammatory agent is selected from hydrocortisone, esters of hydrocortisone and salts thereof.

This invention is directed to a novel formulation for effectively utilizing hydrophobic permeation enhancers for the increased permeability of active agents through the skin or mucosa along with reduction of the lag time between application of the active agent and attainment of a therapeutically effective agent flux, with little or no irritation to the host. The invention is directed to compositions containing a hydrophobic permeation enhancer, which permeation enhancer has been micronized and stabilized in an inert carrier such as water. These compositions can be combined with a biologically active agent to provide enhanced permeability of the active agent to the skin or mucosa.

Formulations with hydrophobic permeation enhancers

An electrotransport delivery device utilizing reservoir buffering at a select pH ranges in order to reduce skin irritation and skin resistance is provided. Cathodic reservoirs are buffered to a pH of less than about 4, preferably to a pH in the range of about 2 to 4, while anodic reservoirs are buffered to a pH above about 4, preferably to a pH in the range of about 4 to 10. Another electrotransport delivery device utilizes a potassium sensor to monitor potassium efflux from the skin. Potassium efflux above a certain predetermined level has been found to be a precursor to skin irritation/erythema. Operation of the device is modified (eg, terminated) when the predetermined potassium efflux level is sensed.
