The present invention is drawn to method for removing unconventional transmissible agents (UCTA) from aqueous protein solution. This method involves treating the aqueous protein solution with simultaneous electrostatic adsorption and hydrophobic chromatography. The method of the present invention is particularly suitable for treating calf serum or an albumin solution.

Method for removing unconventional transmissible agents from a protein solution

The invention relates, as new compounds, to derivatives of tetrameric human haemoglobin corresponding to the covalent coupling of a haemoglobin tetramer and a polycarboxy-substituted compound having at least 4 -COO.sup.- groups, the said polycarboxy-substituted compound effecting an internal crosslinking of the two dimers of haemoglobin and conferring thereon an affinity for oxygen which is compatible with an in vivo use of the conjugate when the natural allosteric effector of haemoglobin is not present. Among carboxy-substituted compounds, special mention is made of benzenehexacarboxylate and benzenetetracarboxylate.

In the method for purifying factor VIII from cryoprecipitate, which is dissolved and then treated with alumina gel, the extract is diluted to a protein concentration not exceeding approximately 5 g/l and subjected to viral inactivation with solvent/detergent, the inactivated extract containing the solvent/detergent is then subjected to chromatography on a weak anion exchange column which is hydrophilic in nature and factor VIII is then eluted with a dissociating buffer.
