Disclosed are compositions of bone precursor cells and methods for their preparation and use. Bone precursor cells are cells which are not hematopoietic and which can differentiate into osteoblasts upon exposure to a bone growth factor and deposit calcium into the extracellular matrix. Such bone precursor cells are useful in the treatment of certain bone related disorders and diseases, such as osteoporosis, or in promoting fracture repair. In addition, methods of differentiating bone precursor cells into osteoblasts, and other diagnostic and even prognostic methods are provided.

Bone precursor cells: compositions and methods

The present invention concerns methods for the ex vivo formation of mammalian bone and subsequent uses of the bone. A critical and distinguishing feature of the present invention are defined tissue culture conditions and factors resulting in the formation of bone cell spheroids. The invention also provides for methods of implanting into subjects the ex vivo formed bone. Also described are methods for genetically altering the bone cell spheroids to affect bone formation, identification of candidate modulators of bone formation, and identification of genes involved in bone formation.

Process for ex vivo formation of mammalian bone and uses thereof

Disclosed are compositions of bone accessory cells and methods for their preparation and use. Bone accessory cells are cells which are not hematopoietic and which can reconstitute the expression of bone proteins by human bone cells and support ex vivo expansion and/or differentiation of these cells. Such bone marrow-derived accessory cells are useful in the treatment of bone disorders and diseases, such as osteoporosis, or in promoting fracture repair. In addition, methods of diffenentiating bone precursor cells into osteoblasts, and other diagnostic and prognostic methods are provided.

The present invention provides compositions of purified cells that are selectively enriched for proliferating hematopoietic progenitor cells. These cells are both CD34 positive and express a galactose-binding surface structure. Methods for the purification and use of these cells, for example, in improving transplantations and reducing graft-versus-host disease also are provided.
