Method and composition for identifying cognate nucleotides in a Sequencing By Binding&#x2122; procedure, wherein one or more labeled nucleotides are detected in ternary complexes but never incorporated. Labeled nucleotides can be incorporable nucleotides that contact preformed blocked primed template nucleic acids. Alternatively, labeled nucleotides are labeled non-incorporable nucleotides. Labeled nucleotides, including labeled non-incorporable nucleotides, can be detected in ternary complexes in the same reaction mixture that incorporates a reversible terminator nucleotide to create a blocked primed template nucleic acid. Detection of ternary complexes can take place in the presence of a catalytic metal ion.

Process for cognate nucleotide detection in a nucleic acid sequencing workflow

Provided herein are methods for analyzing a signature sequence in a nucleic acid sample by rapid sequencing of a target nucleic acid region. The method examines the target nucleic acid directly and minimizes the number of examination steps needed to determine a signature that is characteristic of a genetic feature of the nucleic acid sample.

Methods for determining sequence profiles

Embodiments herein provide for a method of dynamically converting servers and a computer and storage readable medium configured to do the same. In the method a first server enters a server conversion while a second server performs one or more network services. A current physical hardware configuration and logical hardware configuration of a first server and a second server is verified. Possible conversions are calculated of one or more valid logical hardware configurations and one or more valid physical hardware configurations based on the current hardware configurations of the first server and the second server. A valid physical hardware configuration is selected for both servers from the calculated possible conversions. The physical hardware is updated for the conversion of the physical hardware configuration of the first server. The current logical configuration of the first server converts to a valid logical hardware configuration based on the calculated possible conversions.

Methods for generating short tandem repeat (STR) profiles

This invention provides microfluidic devices that comprise a fluidics layer having microfluidic channels and one or more regulating layers that regulate the movement of fluid in the channels. The microfluidic devices can be used to mix one or more fluids. At least a portion of the fluidics layer can be isolated from the regulating layer, for example in the form of a shelf. Such isolated portions can be used as areas in which the temperature of liquids is controlled. Also provided are instruments including thermal control devices into which the microfluidic device is engaged so that the thermal control device controls temperature in the isolated portion, and a movable magnetic assembly including magnets with shields so that a focused magnetic field can be applied to or withdrawn from the isolated portion or any other portion of the microfluidic device. Also provided are methods of mixing fluids. The methods include stacking a plurality of alternating boluses of different liquids in a microfluidic channel, and moving the stacked boluses through the channel. In another method, the boluses are moved into a diaphragm valve having a volume able to accommodate several boluses, and then pumping the liquids out of the valve.

Microfluidic methods

The present invention discloses the integration of programmable microfluidic circuits to achieve practical applications to process biochemical and chemical reactions and to integrate these reactions. In some embodiments workflows for biochemical reactions or chemical workflows are combined. Microvalves such as programmable microfluidic circuit with Y valves and flow through valves are disclosed. In some embodiments microvalves of the present invention are used for mixing fluids, which may be part of an integrated process. These processes include mixing samples and moving reactions to an edge or reservoir for modular microfluidics, use of capture regions, and injection into analytical devices on separate devices. In some embodiments star and nested star designs, or bead capture by change of cross sectional area of a channel in a microvalve are used. Movement of samples between temperature zones are further disclosed using fixed temperature and movement of the samples by micropumps.

Microfluidic and nanofluidic devices, systems, and applications

The invention herein provides improved sample injection systems and related methods to create microfluidic devices with symmetrical channel configurations that can produce relatively large sample volumes. An embodiment of the invention provides microfluidic structures with different geometries that are symmetrical from the perspective of a sample load channel and a sample waste channel, which essentially eliminates issues of time offset and other problems commonly associated with twin-T sample formation techniques. A split-injection approach and related methods of sample plug formation are therefore provided.

Optimized sample injection structures in microfluidic separations

A method and apparatus for managing computer storage devices and updating code in a computing system. An example of the method begins by mirroring at least one logical volume in an original volume group across a first storage device and a second storage device, and then ceasing mirroring the at least one logical volume on the second storage device. The first storage device is kept on-line with the computing system, and information is copied from the first storage device to the second storage device to clone the information from the first storage device. Code is then updated in an alternate volume group on the second storage device, while the computing system is operated with the original volume group on the first storage device. The computing system is then booted from the alternate volume group on the second storage device.
