Bow carriers for use with ATVs and to related methods and systems. In a first illustrative embodiment, a carrier is formed as a box having generally planar front and back sides. At an upper end, the carrier may have an open top and taper to a narrower bottom. The box may be lined with a cushioning material, such as a compressible foam, on either long side with relatively limited space therebetween. The cushioning material may be covered with a suitable textile. The exterior of the box may include hooks and straps or other suitable hardware for attachment to an ATV cargo rack. The open top may be covered by a lid or retractable flap. For use, a bow may be placed in the carrier, into the space between the oppositely mounted cushioning lining. The lining may then be partially compressed to retain the bow securely and safely therein.

Method to protect DNA ends

The present invention provides RecA mutant proteins, having either a single mutation or a double mutation. The RecA mutant proteins are highly proficient in both SSB displacement and steady state binding of DNA in the presence or absence of SSB as compared to the wild-type protein. The single RecA mutant, RecA&#x394;C17, has 17 amino acid residues removed from the carboxyl terminus. The double mutant RecA, RecA&#x394;C17/E38K, combines the 17 amino acid residue C-terminal deletion of RecA&#x394;C17, with a single amino acid change from Glutamate to Lysine at position 38. These RecA mutant proteins are pH sensitive allowing control over formation of products. Hence, methods of using the novel RecA mutants and kits having the RecA mutants as components thereof are also contemplated by the present invention.

A method of introducing exogenous cloned DNA into a bacterial chromosome of a bacteria in which the transposon Tn5 and the FLP recombinase are functional in vivo is disclosed. In one embodiment, the method comprises the steps of: (a) introducing FLP recombination target sites (FRTs) permanently at random locations in a bacterial chromosome using a plasmid vector that contains an FRT within a modified Tn5 transposon, two selectable markers, and a removable replication origin; (b) mapping the FRT introduced into the bacterial chromosome; (c) cloning exogenous DNA into a vector comprising two FRT sites, two selectable markers, and a removable replication origin; (d) removing the replication origin in the vector of step (c); (e) introducing the altered plasmid vector into bacterial cells, wherein the bacteria cells comprise a functional FLP recombinase; and (f) obtaining targeted integrants.

A sculptured optical art form device and the method of making comprising a transparent block having a predetermined index of refraction to provide internal light reflection, said block having a cavity disposed in one wall, the cavity being shaped or carved as a symmetrical one-half of an object that is optically displayed as a whole object in the block. The transparent block may be constructed of an acrylic polymer or glass. The geometrical outside shape of the block is selected to provide an optical view through at least one wall to an observer such that light rays striking the wall having the cavity are internally reflected. The observer looking through the viewing wall will observe a complete, fully formed object optically created by the symmetrical one-half cavity formed in the block wall providing internal optical reflectance. In one embodiment, the object optically created by the cavity appears to be completely embedded in the center of the block.
