Illustrative embodiments of the disclosure are generally directed to barrier-piercing firehose nozzle assemblies suitable for selective application of water and/or other extinguishing liquid to an open fire or piercing a ceiling, floor or other barrier and applying the liquid to a fire concealed above, below or on the other side of the barrier. An illustrative embodiment of the barrier-piercing firehose nozzle assemblies may include a nozzle inlet housing having an inlet housing interior. A spray nozzle may be disposed in fluid communication with the inlet housing interior. A barrier-piercing applicator may be disposed in fluid communication with the inlet housing interior. The barrier-piercing applicator may be selectively deployable between a retracted position and an extended position. At least one discharge opening may be provided in the barrier-piercing applicator. A selector valve may be provided in the inlet housing interior. The selector valve may be selectively deployable in a first valve position in which the selector valve is disposed in fluid communication with the spray nozzle and a second valve position in which the selector valve is disposed in fluid communication with the barrier-piercing applicator.

Methods of preparing pancreatic islets that may comprise pancreatites are provided. The methods may involve subjecting a pancreas and/or pieces thereof to disruption, such as, for example, an enzyme digest, and seeding the recovered cellular product comprising islets in a culture medium comprising at least a detectable amount of endocrine tissue and/or exocrine tissue.

Methods of isolating cellular products, such as pancreatic islets, may be used in diabetes research and therapeutic transplantation. The methods may involve providing a donor tissue having desired cells and undesired cells, perfusing the donor tissue with a perfusion solution, developing edema during perfusion of the donor tissue to form a swelled tissue, and separating the desired cells from undesired cellular material to obtain a cellular product. The methods may also include disrupting the tissue, and separating the desired cells from undesired cellular material to obtain the cellular product. The methods may result in an increased yield of cellular product that retains sufficient functional integrity to be useful as a transplantation resource.

The methods may thereby provide an enzyme-free or reduced-enzyme method of isolating a cellular product that is more consistent, reliable and less toxic than conventional methods. The methods may also yield an optimum quantity of cellular product that retain sufficient functional integrity to be useful as a transplantation resource.

A method and apparatus of plugging and abandoning a well includes using a plug apparatus and installing bentonite elements in an oil well production casing having a bore. The plug apparatus is lowered into the production casing bore to a selected elevation using a slick line or the like. The apparatus has a tool body and slip dies that are movably mounted upon the tool body to move between extended and retracted positions. Afer the tool body is positioned at a selected location within the casing bore, tension is applied to the slick line to move the slip dies to an extended position wherein the slip dies engage the casing and secure the tool body in a desired location. The casing bore is then displaced with water above the tool body. Bentonite elements are then dumped into the water filled wellbore. The bentonite elements are allowed to swell to form a plug. The tool body can be retrieved from the casing bore if desired.

A two stage method of thawing cells from a cryopreserved state includes first warming the cells from a cryopreservation temperature to a transition temperature of at least &minus;30&deg; C. in a first, slow-warming stage by exposing the cells to an atmosphere having a temperature of less than 30&deg; C., and once the cells have obtained the transition temperature, subsequently further warming the cells from the transition temperature by exposing the cells to a temperature of at least 32&deg; C. in a second, rapid-warming stage. After the cells obtain the transition temperature in the first stage, the cells may be equilibrated at the transition temperature for a period of time prior to conducting the second stage warming. The method is particularly useful in warming cryopreserved cells attached to a fixed substrate. A thermal conduction device in association with the cryopreserved cells may also be used to further assist in the warming procedure.
