Novel synthetic catalytic structures or 'synzymes,' e.g., synthetic polypeptides, with catalytic properties are provided. It is believed that these synthetic catalytic structures mimic some of the precise conformational changes necessary for catalytic activities seen in enzymes. The catalytic properties of these synthetic catalytic structures or synzymes can be further improved by the application of controlled external forces, e.g., electric fields.

Devices and techniques are described that involve a combination of multidimensional electrokinetic, dielectrophoretic, electrophoretic and fluidic forces and effects for separating cells, nanovesicles, nanoparticulates and biomarkers (DNA, RNA, antibodies, proteins) in high conductance (ionic) strength biological samples and buffers. In disclosed embodiments, a combination of continuous and/or pulsed dielectrophoretic (DEP) forces, continuous and/or pulsed field DC electrophoretic forces, microelectrophoresis and controlled fluidics are utilized with arrays of electrodes. In particular, the use of chambered DEP devices and of a properly scaled relatively larger electrode array devices that combines fluid, electrophoretic and DEP forces enables both larger and/or clinically relevant volumes of blood, serum, plasma or other samples to be more directly, rapidly and efficiently analyzed. The invention enables the creation of 'seamless' sample-to-answer diagnostic systems and devices. The devices and techniques described can also carry out the assisted self-assembly of molecules, polymers, nanocomponents and mesoscale entities into three dimensional higher order structures.

The concentration step can be carried out by adsorption of the dehybridized probe onto a basic ion exchange material. A displacer sequence having greater binding affinity for the target polynucleotide sequence may be used to dehybridize the bound probe portion for subsequent concentration.

Method for increasing the sensitivity of nucleic acid hybridization

Binding assay methods involving determining the presence of analytes in samples through enzymatic formation of detectable substances in amounts related to the amount of analyte present in the sample and monitoring for the presence of the substances in distinct phases. Methods according to the invention include use of labelled materials which associate with the analyte to be determined or compete with the analyte for association with an added binder. The labelled materials employed include label portions which enzymatically form substances from substrates provided in or existing as a first phase, or, upon enzymatic treatment in a first phase, disassociate into substances capable of existing in or as a second distinct phase. Formation of the detectable substances is monitored by determining the transfer of the substance to a second distinct phase in contact with the first phase or by determining formation of a second distinct phase. The assays are useful in determining human IgG protein in blood samples and other constituents of blood or other biological samples without elaborate instrumentation, allowing for practice outside the clinical laboratory.
