A nozzle for use in delivering a mixture of aerosol propellant and drug formulation. The nozzle includes a drug product inlet configured to receive a mixture of aerosolized propellant and an intranasal dosage form. The inlet is disposed at the proximal end. A nozzle body is secured to the drug product inlet. Two or more channels are disposed within the body. Two or more orifice apertures are disposed at the distal end of the nozzle.

Nozzles for nasal drug delivery

A compound delivery device for delivering a plume derived from a propellant and a drug formulation. The drug formulation is in an intranasal dosage form in the form of powder, suspension, dispersion or liquid. The propelled intranasal dosage form is deposited within the olfactory region of the nasal cavity. The drug deposited within the olfactory region is delivered to the brain avoiding the blood-brain-barrier. Hydrofluoroalkane propellant from a pressurized canister is channeled to a diffuser and drug-containing chamber where the intra-nasal dosage form is aerosolized. The aerosolized intra-nasal dosage form passes through a nozzle thus delivering a plume to the olfactory region of a user's nasal cavity.

Nasal drug delivery device

A method for providing immediate and sustained release of ibuprofen to a subject, including administering, in a single dose, a modified release formulation of ibuprofen including a hydrophilic polymer, ibuprofen in solid dosage form uniformly dispersed in the polymer, and a dissolution additive and an inert formulation additive dispersed in the polymer. The hydrophilic polymer comprises a first hydroxypropyl methylcellulose (HPMC) having a viscosity of about 100 cps and a second HPMC having a viscosity between about 200 cps to about 50,000 cps. The solid dosage form is a monolithic tablet that demonstrates a mean ibuprofen plasma concentration in a subject greater than or equal to 6.4 &#x3bc;g/ml within two hours of administration, and also demonstrates a mean ibuprofen plasma concentration in a subject greater than or equal to 6.4 &#x3bc;g/ml for at least 8 hours after administration.

Method for providing modified release of ibuprofen

A composition is described having improved oral permeability of polar agents such as neuraminidase inhibitors. The composition includes one or more polar agents and one or more permeability enhancers such that the composition increases the amount of the polar agent capable of being transported across a Caco-2 cell membrane by at least 150% relative to the amount capable of being transported across the Caco-2 Cell membrane in the absence of the permeability enhancer. Oral dosage forms including the composition, and methods of treating or preventing influenza infection are also provided.

Formulations for enhanced bioavailability of orally administered polar agents

The present invention is a solid dosage form for oral administration of ibuprofen comprising a modified release formulation of ibuprofen which provides an immediate burst effect and thereafter a sustained release of sufficient ibuprofen to maintain blood levels at least 6.4 &#x3bc;g/ml over an extended period of at least 8 hours following administering of a single dose.

A controlled release delivery system composition and method applied to humans and animals, for oral, anal or vaginal administration of a biological component is disclosed. Preferably, a bacterium is delivered, and more preferably the bacterium is probiotic in nature, however, the biological component is not limited to the bacterium.

Delivery system for biological component

The invention relates to a controlled release formulation for a probiotic. The formulation contains a hydrophilic agent, an electrolytic agent and a polysaccharide and is in the form of a monolithic tablet for oral delivery to the intestinal system. The probiotic can be lactic acid bacteria.

A method of forming a tablet includes the steps of pre-blending an active pharmaceutical ingredient susceptible to tackiness and a blending additive with a first mixing effort to form a pre-blend mixture, wherein the first mixing effort and a second mixing effort, resulting from mixing at least one excipient with the pre-blend mixture, form a blend suitable for direct compression and compressing the blend to form the tablet. One way of achieving the first mixing effort is to pre-blend for an extended period of time. The method allows for directly compressing the blend without the need for a granulation step or roller compression. One such active pharmaceutical ingredient susceptible to tackiness is ibuprofen.
