Methods are described for generating autologous tissue grafts, including generating grafts at the point of care, which include isolated cell populations that are enriched with stem cells and are mixed with biological fillers including hyaluronic acid and derivatives thereof. The hyaluronic acid localizes the cells to a desired injection site and stimulates collagen production thus enhancing the viability and the longevity of the graft.

Methods for obtaining target cells

A culture media, media supplement, or soluble matrix for cryopreservation or enhanced regenerative cell growth in culture and maintenance of multi-lineage differentiation potentiation. The inventive culture media, media supplement, or soluble matrix comprises a GAG composition comprising a sulfated GAG, such as chondroitin sulfate. A soluble matrix, a cell administration package or kit comprising the soluble matrix and a device for cell administration, and a method of use thereof, for administration of regenerative cells for treating a joint disease or other weakened or damaged tissue comprising the specified GAG compositions are further provided.

Media for culturing, preserving, and administering regenerative cells

A unitary apparatus for isolating cells from adipose tissue including a lipid separation processor with a dispersing head equipped with a plurality of ports and a digestion chamber for dissociation of the constituent cells disposed in adipose tissue. The lipid separating apparatus is useful for the separation of lipids and adipocytes from a mixed cell population. A cell seeding chamber may be attached to the cell isolation apparatus. The components of the apparatus may be packaged in modular kit form.
