A method for multiplexed detection and quantification of analytes by reacting them with probe molecules attached to specific and identifiable carriers. These carriers can be of different size, shape, color, and composition. Different probe molecules are attached to different types of carriers prior to analysis. After the reaction takes place, the carriers can be automatically analyzed. This invention obviates cumbersome instruments used for the deposition of probe molecules in geometrically defined arrays. In the present invention, the analytes are identified by their association with the defined carrier, and not (or not only) by their position. Moreover, the use of carriers provides a more homogenous and reproducible representation for probe molecules and reaction products than two-dimensional imprinted arrays or DNA chips.

Systems and methods of conducting multiplexed experiments

A method is provided for measuring an analyte in a sample comprising adding substantially transparent particles to a sample in solution or suspension, said particles having an affinity for said analyte; fractionating the particles from the solution or suspension to form a particle-rich fraction and a substantially particle-free fraction; optically reading the particle-rich fraction at a first and a second wavelength; optically reading the substantially particle-free fraction at at least the first wavelength; and correlating the readings through the particle-rich fraction and the substantially particle-free fraction of the sample, with similar measurements in a particle-containing 'blank' to obtain a quantitative determination of the analyte originally present in the sample.

Two-phase optical assay

Methods and apparatus for separating red blood cells by density are described. Older red blood cells are more dense than younger ones. Certain changes in the physiology are recorded in changes in hemoglobin or the red blood cells. By assaying different aged red blood cells, one can determine the historical physiological changes over a period of many weeks. Improved separation of red blood cells is accomplished by using rigid capillary tubes having an inner surface which augments the density equilibrium of red blood cells and/or incrementally increasing the centrifugation forces, as well as by chemically treating red blood cells to improve their deformability.

Method and procedure for preparing red blood fractions

A method for isolating and enriching rare cells, including fetal nucleated erythrocytes from a peripheral blood sample is described. The method includes two centrifugation steps, the first a bulk separation step to enrich erythrocytes from other blood components. The second centrifugation comprises a colloidal density gradient medium dispersed in a hypertonic meltable gel. In one aspect of the invention, maternal erythrocytes may be hemolyzed prior to the second density gradient centrifugation to provide additional enrichment for fetal nucleated erythrocytes.

Method for separating rare cells from a population of cells

A centrifuge tube and adaptor apparatus is provided which facilitates separation of biological materials and permits easy extraction of a fraction after centrifugation. The tube is a deformable tube with a wide upper chamber and a narrowed lower portion. The tube is supported within the centrifuge rotor or within another container within the centrifuge by a liquid support medium, which surrounds and supports the narrow portion of the tube, and thus prevents the tube from collapsing during high speed centrifugations.
