The present disclosure relates generally to methods and compositions useful in cell and tissue biology and therapeutics. In particular, an in vitro method for differentiating pluripotent stem cells into KDRNCAMAPLNRmesoderm cells and/or SSEA5KDRNCAMAPLNRmesoderm cells is provided. The disclosed mesoderm cells may be used to generate blood vessels in vivo and/or further differentiated in vitro into endothelial colony forming cell-like cells (ECFC-like cells). Purified human cell populations of KDRNCAMAPLNRmesoderm cells and ECFC-like cells are provided. Test agent screening and therapeutic methods for using the cell populations of the present disclosure are provided.

Method for generating mesoderm and/or endothelial colony forming cell-like cells having in vivo blood vessel forming capacity

Materials and methods are disclosed for controlling vasculogenesis using building blocks of a collagen matrix and endothelial colony forming cells (ECFC). The building blocks may be isolated by fractionating an acid soluble Type I collagen. The building blocks comprising monomers and/or oligomers may be recombined in desired ratios to alter the matrix microenvironment and to influence ECFC behavior.

Materials and methods for controlling vasculogenesis from endothelial colony forming cells

A hierarchy of endothelial colony forming cells (EPCs) was identified from mammalian cord blood, umbilical vein and aorta. A newly isolated cell named high proliferative potential&#x2014;endothelial colony forming cell (HPP-ECFC) was isolated and characterized. Single cell assays were developed that test the proliferative and clonogenic potential of endothelial cells derived from cord blood, or from HUVECs and HAECs. EPCs were found to reside in vessel walls. Use of a feeder layer of cells derived from high proliferative potential-endothelial colony forming cells (HPP-ECPCS) from human umbilical cord blood, stimulates growth and survival of repopulating hematopoietic stem and progenitor cells. Stimulation of growth and survival was determined by increased numbers of progenitor cells in in vitro cultures and increased levels of human cell engraftment in the NOD/SCID immunodeficient mouse transplant system.

Isolation, expansion and use of clonogenic endothelial progenitor cells

The present disclosure relates generally to methods and compositions useful in cell and tissue biology and therapeutics. In particular, an in vitro method for differentiating pluripotent cells into endothelial colony forming cell-like cells (ECFC-like cells) is provided. A purified human cell population of NRP-1+CD31+ ECFC-like cells is provided, wherein at least some of the cells in the population have a high proliferation potential. Therapeutic and test agent screening methods for using the cell populations of the present disclosure are provided.

Collagen based-matrices and methods of their use are described. More particularly, collagen-based matrices for differentiating stem cells and progenitor cells, and for producing and isolating blood vessels and vascularized graft constructs are described.

Collagen-based matrices with stem cells

The roof jacking seat system is inclusive of a seat pivotally oriented with respect to a wheeled frame such that said seat can adjust in accommodation of different roof pitches. The system is for the support of an end user atop of said roof during roof shingle removal processes. The frame includes a winching means having a cable and hook for engagement onto a roof ridge such that the entire system may be driven up or down a side of said roof. The frame includes a plurality of wheels in parallel orientation thereby enabling movement along a single axis. Pivoting means enables adjustment of the seat at different angles with respect to the frame. The frame may include along a distal end, a footrest for use by the end user.
