Provided are methods and compositions for treating an individual who has a medulloblastoma. Methods for treating an individual who has a medulloblastoma tumor can include a step of administering to the individual, at a dose sufficient to reduce the size and/or growth rate of the medulloblastoma tumor, a composition that includes a casein kinase II (CK2) inhibitor (e.g., a CK2-selective inhibitor such as CX-4945). In some cases, the medulloblastoma tumor is a hedgehog-dependent medulloblastoma tumor. In some cases, the medulloblastoma tumor is a hedgehog-independent medulloblastoma tumor. In some cases, the medulloblastoma tumor is smoothened inhibitor-resistant (SMO inhibitor-resistant). In some cases, the medulloblastoma is resistant to treatment with 4,5,6,7-tetrabromo-2H-benzotriazole (TBB). In some cases, the dose is sufficient to cause long term regression of the medulloblastoma tumor, and in some cases, the dose is sufficient to increase the chance of survival of the individual.

Compositions and methods for treating medulloblastoma

Methods for isolating patched genes, including the mouse and human patched genes, as well as invertebrate patched genes and sequences, are provided. Decreased expression of patched is associated with the occurrence of human cancers, particularly basal cell carcinomas of the skin. The patched and hedgehog genes are useful in creating transgenic animal models for these human cancers. The patched nucleic acid compositions find use in identifying homologous or related proteins and the DNA sequences encoding such proteins; in producing compositions that modulate the expression or function of the protein; and in studying associated physiological pathways. In addition, modulation of the gene activity in vivo is used for prophylactic and therapeutic purposes, such as treatment of cancer, identification of cell type based on expression, and the like. The DNA is further used as a diagnostic for a genetic predisposition to cancer, and to identify specific cancers having mutations in this gene.

Methods are provided for identifying agents that decrease hedgehog signal transduction for ameliorating an effect of loss of function of a patched gene in a mammalian cell.

An automated particle sorter having a fluid flow path, which places single biological particles in an optical cuvette. An exciting light irradiation system having a light source emits a source of light through the cuvette. The light excites a fluorescent substance present on the particle, and the emitted light is detected by a light detection apparatus containing at least two detection elements for measuring the fluorescence emitted from the fluorescent substance. A light separation element separates the fluorescence from the exciting light. A data processor compares the signal received from the fluorescent light; and from the background autofluorescent signal, and according to pre-set parameters, controls the position of a collection conduit between two set points.
