A mixture of components is flowed through a binding channel region comprising a component-binding moiety, thereby binding at least a portion of a component of interest. The mixture is then flowed through a separation channel region that includes a buffer comprising a detergent, resulting in separated components. Diluent is mixed with the separated components, diluting the detergent, and the separated components are detected. The component of interest is released from the component-binding moiety and flowed through the separation channel region. Diluent is mixed with the released component of interest, diluting the detergent, and the released component of interest is detected.

Western blot by incorporating an affinity purification zone

An upstream affinity purification region is used to bind one or more component of interest in a mixture of components prior to separating the mixture of components. Detection of the separated components and a released component of interest provide identification of the component of interest. In addition, post separation dilution is optionally used to improve detection of the mixture of components and the released component of interest. Microfluidic devices and systems suitable for performing such analyses are also provided.

Methods of detecting a component of interest, such as a protein, in a microfluidic system are provided. The methods include the use of a component-binding moiety specific to the component of interest, such as an antibody, to detect the component of interest. Also included are microfluidic devices and integrated systems for performing such assays, including devices utilizing flowable or fixed particle sets.

Microfluidic analytic detection assays, devices, and integrated systems

This invention provides a method of recovering and purifying chymosin from fermentation used for microbial production of chymosin. The fermentation mixture is filtered to provide a filtrate containing the chymosin. The filtrate is first prepared by either adjusting it to a low pH, such as about 2 and/or adjusting the salt concentration of the filtrate to at least about 2M. The aqueous filtrate mixture is contacted with a phenyl-sepharose resin, which selectively binds the chymosin from the filtrate. The filtrate remaining after the contact with the phenyl-sepharose resin is discarded, and the phenyl-sepharose resin column is eluted with water or dilute salt solution to obtain substantially pure chymosin in a single elution step.
