The disclosure provides CARs (CARs) that specifically bind to CD70. The disclosure further relates to engineered immune cells comprising such CARs, CAR-encoding nucleic acids, and methods of making such CARs, engineered immune cells, and nucleic acids. The disclosure further relates to therapeutic methods for use of these CARs and engineered immune cells comprising these CARs for the treatment of a condition associated with malignant cells expressing CD70 (e.g., cancer).

Chimeric antigen receptors targeting CD70

The invention provides Chimeric Antigen Receptors (CARs) that specifically bind to EGFRvIII (Epidermal Growth Factor Receptor Variant III). The invention further relates to engineered immune cells comprising such CARs, CAR-encoding nucleic acids, and methods of making thereof, engineered immune cells, and nucleic acids. The invention further relates to therapeutic methods for using these CARs and engineered immune cells for the treatment of EGFRvIII-mediated pathologies, including cancers such as glioblastoma.

Methods of developing genetically engineered immune cells for immunotherapy, which can be endowed with Chimeric Antigen Receptors targeting an antigen marker that is common to both the pathological cells and said CD38 immune by the fact that the genes encoding said markers are inactivated in said immune cells by a rare cutting endonuclease such as TALEN, Cas9 or argonaute.
