A portion of the wetted surfaces are coated with an alkylsilyl coating having the Formula I: R, R, R, R, R, and Rare each independently selected from (C-C)alkoxy, —NH(C-C)alkyl, —N((C-C)alkyl), OH, OR, and halo. Rrepresents a point of attachment to the interior surfaces of the fluidic system. At least one of R, R, R, R, R, and Ris OR. X is (C-C)alkyl, —O[(CH)O]—, —(C-C)[NH(CO)NH(C-C)]—, or —(C-C)[alkylphenyl(C-C)alkyl]-.

Use of vapor deposition coated flow paths for improved analytical analysis

A device for separating analytes is disclosed. The device has a sample injector, sample injection needle, sample reservoir container in communication with the sample injector, chromatography column downstream of the sample injector, and fluid conduits connecting the sample injector and the column. The interior surfaces of the fluid conduits, sample injector, sample reservoir container, and column form a flow path having wetted surfaces. A portion of the wetted surfaces of the flow path are coated with an alkylsilyl coating that is inert to at least one of the analytes. The alkylsilyl coating has the Formula I: R, R, R, R, R, and Rare each independently selected from (C-C)alkoxy, —NH(C-C)alkyl, —N((C-C)alkyl), OH, OR, and halo. Use of vapor deposition coated flow paths for improved chromatography of metal interacting analytes

The present disclosure relates to a method of reducing degradation of sample components in a liquid chromatography system. The method utilizes a masked metal frit to prevent or reduce metal surfaces from becoming catalytically active. The masked metal frit is a metal based frit that includes a coating on its exterior surfaces to mask or prevent contact between the organic solvents (and/or any analyte or other related solvent) and the underlying metal. The coating is a vapor deposited inorganic-organic hybrid coating, such as a vapor deposited C2 coating.

Chromatographic systems for size exclusion chromatography (SEC) are provided that comprise an inlet, an outlet, an analytic column having a first interior volume that has a first length and a first cross-sectional area normal to the first length, the first interior volume containing a first stationary phase, and a guard column having a second interior volume that has a second length and a second cross-sectional area normal to the second length, the second interior volume containing a second stationary phase. The inlet is in fluid communication with the guard column, the guard column is in fluid communication with the analytic column, and the analytic column is in fluid communication with the outlet. Moreover, the second length is smaller than the first length, and the second cross-sectional area is smaller than the first cross-sectional area.

The present disclosure is directed to methods of characterizing a system containing a chromatographic column. The methods can include introducing a sample comprising a positive control and a negative control to the system containing a chromatographic column, wherein the positive control is a sensitive probe that interacts with the system and the negative control is substantially non-interacting with the system; after passing the sample through the chromatographic column, detecting the positive control and the negative control; and determining system suitability by comparing the amount of detected positive control to negative control. In some embodiments, determining system suitability (e.g., inertness of sample to the system) is accomplished by determining a ratio of detected positive control to negative control.

The present disclosure relates to the determination of analytes in a sample using chromatography. The present disclosure provides methods of separating an analyte from a sample. The method includes introducing a sample comprising the analytes into a chromatographic system. The chromatographic system has a flow path disposed in an interior of the chromatographic system, at least a portion of the flow path having an active coating, and a chromatographic column having a stationary phase material in an interior of the chromatographic column that facilitates separation of the analytes in the sample through interaction with at least one analyte in the sample. The active coating is selected to interact with at least one analyte in the sample through (1) a repulsive force, (2) a secondary interaction, or (3) a retention mechanism distinct from the interaction with the stationary phase material.

Coated flow path components for chromatographic effects

The present disclosure is directed to methods for evaluating system inertness, such as the inertness of a LC or other fluidic system. Some methods are directed to tests wherein the column has been removed prior to injecting a sample including a positive (e.g., metal reacting moiety) control into the system. Some methods can include: (1) repeatedly injecting the sample into a system, the system comprising: fluidic paths wherein interior surfaces of the fluidic paths define wetted surfaces, and wherein at least a portion of the wetted surfaces of the fluidic flow path are coated with an inert coating, wherein the inert coating is inert to at least one analyte in the sample; (2) detecting a value associated with the positive control; and (3) analyzing values associated with the detected positive control to determine system inertness.
