Methods and the resulting structures, like valve devices (e.g. heart or vessel) are provided made by electrospinning single shapes on mandrels with complex surface shapes. These single shapes are then shaped into valves with a plurality of leaflets. Three levels of complexity of shapes are described: 1) conical shapes, 2) a combination of conical and cylindrical shapes, and 3) a conical and/or cylindrical shape which has further complexity by one or more three-dimensional shapes. Heart valves resulting from these complex electrospun shaped mandrels have better mobility dynamics compared to heart valves electrospun on solely cylindrical mandrels.

Electrospun heart valves

A cardiovascular graft is provided with highly reduced thrombogenicity. The cardiovascular graft is an electrospun non-woven mesh produced from supramolecular polymers with large diameter fibers. The cardiovascular graft can be implemented as a vascular graft into the human body to allow vascular bypass/reconstruction, or repeated venous access for dialysis treatment, as well as other disorders of small-diameter blood vessels.

Bioabsorbable medical devices such as vascular closures, mitral chorea replacements, and mitral leaflet extensions are provided.

A method of making a medical implant is provided by electrospinning a polymer solution to form a preform around a mandrel. The formed preform distinguishes an inner surface and an outer surface. The formed preform is removed from the mandrel and flipped inside-out resulting in the inner surface of the formed preform becoming the outer surface of the inside-out flipped preform, and the outer surface of the formed preform becoming the inner surface of the inside-out flipped preform. At least part of the inside-out flipped preform forms the medical implant such as e.g. an artificial heart valve, an artificial leaflet, an artificial graft, or an artificial vessel. The products made according to the method of this invention greatly improve the performance and durability of the medical implant.

The disclosure lies in the field of regenerative medicine and relates to an implant having a matrix material, and a method for manufacturing an implant having matrix material. The disclosure further relates to a crimped implant.

Implant

A biodegradable cardiovascular implant is provided for growing cardiovascular tissue in a patient. The implant distinguishes an electro-spun network with supramolecular compounds having hard-blocks covalently bonded with soft-blocks resulting in much enhanced durability and fatigue resistance, while maintaining the effectiveness as a cardiovascular implant.

The present disclosure relates to in situ tissue engineering, more specifically in situ vascular tissue engineering with the aim of providing a tissue structure which can be used e.g. as a substitute blood vessel or as a blood vessel functioning as cannulation site in hemodialysis. In particular, the disclosure involves tissue structure formation around a subcutaneously implanted synthetic rod. In addition, the disclosure involves a method for producing said synthetic rod.

In situ tissue engineering

Endoluminal and other as implantable prostheses are fabricated in electrospinning apparatus including a target and an applicator. A solution comprising a polymer and a solvent is directed to the target with a first electrical potential between the target and the applicator to produce a first set of fibers. The same or another solution is continued to be delivered through the applicator onto the target while applying a second electrical potential to produce a second set of fibers having a second solvent fraction, and the same or different solution may be delivered while applying a third potential difference to produce a laminated structure having at least three layers. By properly controlling the electrical potentials and solvent fractions, an adhesive layer can be formed to serve a glue or adhesive between the inner and outer layers, and a stent or other scaffold may be positioned between the inner and outer layers to form a covered stent or graft.

Methods for electrospin coating and laminating of endoluminal prostheses

The invention relates a multilayer preform obtained by electro-spinning, which preform is suitable as a scaffold for a prosthesis, which preform comprises at least one layer of microfibers and at least one layer of nanofibers, wherein the pore size of the at least one layer of microfibers is in the range of 1-300 micrometer and in that the pore size of the at least one layer of nanofibers is in the range of 1-300 micrometer. The present invention also relates to a method of producing said preform. The present invention also relates to the use of the present preform as a substrate for growing human or animal tissue thereon. The present invention furthermore relates to a method for growing human or animal tissue on a substrate, wherein the present preform is used as the substrate.
