Compositions and methods for binding to a target sequence of interest are provided. Compositions include fusion proteins between DNA binding proteins or protein domains and nucleic acid modifying proteins or protein domains. The compositions find use in cleaving or modifying a target sequence of interest, visualization of a target sequence of interest, and modifying the expression of a sequence of interest. Compositions comprise RNA-guided nuclease polypeptides, CRISPR RNAs, trans-activating CRISPR RNAs, guide RNAs, deaminases, and nucleic acid molecules encoding the same. Vectors and host cells comprising the nucleic acid molecules are also provided. Further provided are CRISPR systems for binding a target sequence of interest, wherein the CRISPR system comprises an RNA-guided nuclease polypeptide and one or more guide RNAs. Also provided are deaminases which may be fused to a DNA-binding polypeptide and may be useful for gene editing.

Polypeptides useful for gene editing and methods of use

An aerial vehicle may have a fuselage, a first wing member, and a second wing member. The fuselage may have a nose end and a tail end. The first wing member may extend from the fuselage and comprise a first drive motor coupled to the first rotor. The second wing member may also extend from the fuselage substantially opposite the first wing member and may comprise a second drive motor coupled to a second rotor. A first motor may be coupled to rotate the first wing member and the first rotor about a first axis substantially perpendicular to a fuselage axis extending from the nose end to the tail end. A second motor may be coupled to rotate the second wing member and the second rotor about a second axis substantially perpendicular to the fuselage axis. A controller circuit may be configured to differentially actuate the first motor and the second motor.

Aerial vehicle with differential control mechanisms

A vertical landing and take-off aircraft VTOL transitions from a vertical takeoff state to a cruise state where the vertical takeoff state uses propellers to generate lift and the cruise state uses wings to generate lift. The aircraft has an M-wing configuration with propellers located on the wingtip nacelles, wing booms, and tail boom. The wing boom and/or the tail boom can include boom control effectors. Hinged control surfaces on the wings, tail boom, and tail tilt during takeoff and landing to yaw the vehicle. The boom control effectors, cruise propellers, stacked propellers, and control surfaces can have different positions during different modes of operation in order to control aircraft movement and mitigate noise generated by the aircraft.

VTOL M-wing configuration

A request for transport services that identifies a rider, an origin, and a destination is received from a client device. Eligibility of the request to be serviced by a vertical take-off and landing (VTOL) aircraft is determined based on the origin and the destination. A transportation system determines a first and a second hub for a leg of the transport request serviced by the VTOL aircraft and calculates a set of candidate routes from the first hub to the second hub. A provisioned route is selected from among the set of candidate routes based on network and environmental parameters and objectives including pre-determined acceptable noise levels, weather, and the presence and planned routes of other VTOL aircrafts along each of the candidate routes.

Dynamic aircraft routing

An aerial vehicle includes a fuselage supporting a pair of wings, with each of wings having a pair of booms attached thereto. A tilt rotor is positioned at each of the forward ends of each of the booms, to provide the aerial vehicle with at least four tilt rotors. A fixed rotor is positioned at and secured to the aft (or trailing) ends of each of the booms, to provide the aerial vehicle with at least four fixed rotors.

An aircraft can include a stacked propeller to generate lift during assent and descent. The stacked propeller includes a first propeller and a second propeller that co-rotate about an axis of rotation. In one embodiment, the blades are coupled to a rotor mast that contains an internal cavity. In one mode of operation, the first propeller and/or the second propeller can be stored in the internal cavity in order to reduce drag during flight. The aircraft can include one or more stacked propellers, such as a port propeller and a starboard propeller, which rotate in opposite directions during one or more modes of flight.

The mobile communication device comprises a processor, at least one cellular radio transceiver, a non-transitory memory, and an ad hoc service upgrade application. When executed by the processor, the application monitors wireless cellular communication on a network via the at least one cellular radio transceiver, detects a significant delay in the communication, generates a prompt on a GUI, the prompt comprising a plurality of options for an ad hoc temporary upgraded QoS, where each option is associated with a time period. The application further, based on a user input, requests a preferred roaming list (PRL) associated with the temporary upgraded QoS, receives the PRL, wherein the PRL is associated with providing the upgraded QoS on an allocated spectrum, activates the received PRL, and upon expiration of the time period, deactivates the received PRL.

Methods, systems, apparatus, and computer program products related to a digital business management system for business transactions are provided. In an example embodiment, the digital business management system includes receiving a transaction data object generated as a part of a transaction event executed via an external commerce channel to be posted to a ledger, wherein the transaction data object comprises one or more metadata tags classifying the transaction event. The system further comprises generating an immutable ledger entry, parsing the transaction data object to identify the one or more metadata tags for storage in association with the transaction event, identifying a match between the one or more metadata tags received from the external commerce channel and one or more master metadata tags stored in the ledger and associated with a data object profile, and updating the data object profile by associating the immutable ledger entry with the data object profile.

Systems and methods for processing, securing, and communicating industrial commerce transactions

A device is provided for enclosing an extremity of a patient for container efficient wound cleaning and for integrating draping prior to surgery. A loose-fitting flexible container with a flexible seal is provided for enclosing the extremity and treating the extremity in a controlled fashion to reduce or eliminate spillage and unwanted dispersion of antiseptic solution. Sealed to the flexible container is a vacuum packed, folded and rolled sterile surgical drape. The combination of a flexible container and a sterile pre-positioned surgical drape in a single device provides the advantage of controlled decontamination of the skin or wound while simultaneously providing the advantage of pre-positioning a surgical drape. The use of the device also provides the advantages of reduced operating room fires and biohazard dispersion risk and reduced cost by reduction of total operating room time per procedure.

Apparatus and method for deploying a preoperative skin disinfection device with integrated drape

A device used for applying an antiseptic preparation, such as an antibiotic solution or water, or all other substance deemed necessary in treating a wound prior to arrival at a hospital or other treatment facility or for continuing wound therapy and maintenance while hospitalized, in an out-patient facility or at home. A loose-fitting bag is provided which encloses the wound by securing the open end(s) to the patient by closing means. The bag further includes ports or resealable flaps that allow access to the wound for treatment and cleaning while containing all fluids resulting therefrom. A high friction scrub surface is provided in the bag. A method and apparatus are also provided for wound maintenance with a lavage system.

Apparatus and method for deploying a surgical preparation

A robocall management system (RMS) comprises a service switching point switch, a service control point, and a robocall confirmation system, that all cooperate in processing incoming calls to a subscriber. If the call is not a robocall, the call will be delivered to the subscriber as normal. If the call is, or may be, a robocall, then call will be diverted to the robocall confirmation system, which will determine whether the caller is a live person or a computer playing an announcement. If the caller is a live person, then the call is offered to the subscriber. If the caller is a robocaller, then the robocall announcement is recorded by the robocall confirmation system and made available to the subscriber should they choose to listen to it. Various algorithms may be employed along with external data to ascertain a likelihood of whether the call is a robocall.

Robocall management system

A device used for applying an antiseptic preparation to a surgical site of a patient prior to surgery outside of an operating theater. A loose-fitting bag is provided which encloses the surgical site by securing the open end(s) to the patient by closing means. A high friction scrub surface is provided in the bag. A method and apparatus are also provided for releasing antiseptic into the interior of the bag. The method provides that the surgical site can be scrubbed within the bag so as to properly prepare the skin for surgery. A temperature indicator for the antiseptic preparation is also provided.

An occupiable building and a method for constructing an occupiable space in a structural frame building are disclosed. In an embodiment, an occupiable building includes a structural frame defining a footprint of the occupiable building, at least one lower deck structure located within the footprint of the structural frame and supported by the structural frame, at least one upper deck structure located within the footprint of the structural frame and supported by the structural frame, and an interior partition system installed between the lower deck structure and the upper deck structure to define an occupiable space, the occupiable space having a ceiling. The interior partition system includes first king post assemblies, a lower panel, and an upper panel. The lower panel is fastened to the king post assemblies and to the upper panel. The upper panel is fastened to the king post assemblies and to upper panel.
