A distributed system processing a publisher's streaming data. The distributed system comprises multiple workers and publisher data stores, each publisher data store dedicated to a worker and a publisher. A sampling ratio (the fraction of data items for storage in the publisher's data store) is selected by a publisher data store's worker based on historical information. At least two workers select different sampling ratios. Data items representing an interaction between an entity and the publisher are received. Each data item is assigned to a worker for processing. A hash function is applied to the data item's identifier, resulting in a key value falling within the hash function's range. The scope of the publisher's data store is equal to the hash function's range multiplied by the sampling ratio of the publisher's data store. A data item with a key value within the scope of the publisher's data store is stored therein.

Managing a distributed system processing a publisher's streaming data

Embodiments are directed to generating and managing an advertising campaign based on a third-party sales listing. In particular, an identifier associated with a sales listing posted over a third-party service from a user of the third-party service is received. Sales listing data for the sales listing is retrieved from the third-party service using the identifier associated with the sales listing. Thereafter, an advertising campaign is automatically generated based at least in part on the retrieved sales listing data. In one aspect, generation of the advertising campaign includes generating an advertising creative using the retrieved sales listing data. Subsequently, bidding on an advertising opportunity in an auction is performed according to the generated advertising campaign. Responsive to winning the auction, an advertisement based on the advertising creative is provided to a content provider associated with the advertising opportunity.

Disclosed herein are methods and compositions for the treatment of diseases of the CNS with nucleic acid nanoparticles. Compositions are also disclosed herein that utilize nucleic acid nanoparticles to treat conditions such as Parkinson's Disease. Furthermore, methods of intranasally administering the compacted nucleic acid nanoparticles for therapeutic purposes in the brain are disclosed.

Methods for delivery to the central nervous system of nucleic acid nanoparticles to treat central nervous system disorders

Efficient and prolonged hCFTR expression is one of the major obstacles for cystic fibrosis lung therapy. hCFTR mRNA expression levels depend on eukaryotic expression cassette components, prokaryotic backbone elements, and the gene transfer method may also influence transcriptional silencing mechanisms. A codon-optimized and CpG-reduced human CFTR gene (CO-CFTR) was made. Various vector modifications were tested to facilitate extended duration of CO-CFTR expression. Insertion of an extended 3'BGH transcribed sequence (712 bp) in an inverted orientation produced prolonged expression of CO-CFTR expression at biologically relevant levels. Further studies revealed that prolonged CO-CFTR expression is dependant on the orientation of the extended BGH 3&#x2032; BGH transcribed sequence and its transcription, is not specific to the UbC promoter, and is less dependent on other vector backbone elements.

Long-term in vivo transgene expression

Counterions of polycations used to compact nucleic acids profoundly affect shape and stability of particles formed. Shape is associated with differential serum nuclease resistance and colloidal stability. A surrogate for determining such properties that is easy to measure is the turbidity parameter. Shape also affects the suitability and efficacy of compacted nucleic acid complexes for transfecting cells by various routes into a mammalian body. Lyophilizable and enhanced compacted nucleic acids

An automated nucleic acid compaction device for analyzing and compacting a nucleic acid complex into unimolecular nucleic acid particles. Broadly, the device includes a container support and agitation system; a measuring and testing system; and a dispensing system; all controlled by a control system. The control system controls the support and agitation system and the dispensing system based either on a predetermined formulation or by analysis of feedback data provided by the measuring and testing system. In a preferred embodiment, the device is a real-time measuring and mixing instrument operating in a closed loop system. The preferred embodiment also comprises a monitoring system including a submersible probe which is positioned in the batch solution to provide data to a controller. Once a desired level of nucleic acid compaction is reached, as detected by the monitoring system, the controller stops the dispensing and mixing agitating systems.

Method of nucleic acid compaction

Episomal plasmids containing a papovavirus origin of replication and a papovavirus large T antigen mutant form are shown to replicate episomally in human cells, and yield levels of gene expression proportional to their episomal copy number. In conjunction with liposomal or receptor-mediated delivery systems, papovavirus-derived episomal plasmids provide an alternative vector for gene therapy, particularly when utilizing strategies requiring high levels of gene expression.

Automated nucleic acid compaction device

Episomal plasmids containing a papovavirus origin of replication and a papovavirus large T antigen mutant form are shown to replicate episomally in human cells, and yield levels of gene expression proportional to their episomal copy number. Candidate oncogenes and anti-oncogenes can be identified by transfecting the episomal plasmids containing a candidate oncogene or anti-oncogene into non-tumorigenic cells and screening for a tumorigenic phenotype.
