The present invention is related to variants of fungal serine protease enzyme, which have serine protease activity ofprotease. Also disclosed are isolated nucleic acid molecules, comprising polynucleotide sequences which encode variants of fungal serine protease enzyme, nucleic acid sequences encoding said protease variants, a host cell and a process of producing polypeptides having serine protease activity. Said protease variants are useful as enzyme preparations applicable in detergent compositions and for treating fibers, wool, hair, leather, or silk, for treating food or feed, or for any applications involving modification, degradation or removal of proteinaceous material.

Protease enzyme variants and uses thereof

The present description is related to the field of protein production. It introduces novel host cells with low protease activity, a novel protease regulator, its use in expression systems and protein production, and a method of producing host cells for protein production.

Methods for controlling protease production

The present invention is related to a fungal serine protease enzyme, which said enzyme has serine protease activity and comprises an amino acid sequence ofALKO4122 mature protease as defined in SEQ ID NO:18 or an amino acid sequence having at least 66% identity to the amino acid sequence of SEQ ID NO:18. Also disclosed is an isolated nucleic acid molecule, comprising a polynucleotide sequence which encodes a fungal serine protease enzyme, nucleic acid sequences encoding said protease, a host cell and a process of producing a polypeptide having serine protease activity. Protease enzyme and uses thereof

The invention is related to a method and test kits for quantitative determination of polynucleotide amounts present in a sample. The test kit comprises organized pools with polynucleotide probes having distinct sizes and optionally provided with tracer tags or primer tags. The probes are allowed to hybridize with affinity tagged analyte polynucleotides from the sample. The result is hybrids, which can be recovered on a separation aiding tool provided with the pair of the affinity tag. After the quantitative release of the probes, the probes are either directly recorded, or if primer tagged, they are amplified and optionally provided with a tracer tag before recording. The invention provides a sensitive and quantitative determination of the amount polynucleotides present in a cell or tissue sample and allows a quantitative assessment of variations in the amounts of polynucleotides as a response to inherent changes or due to external stimuli.

Method for determining amounts of polynucleotide sequences present in cell or tissue samples

The present invention relates to fungal serine protease variants, which comprise an amino acid substitution of valine at position 208 of the parentFe_RF6318 serine protease, wherein the position of the substitution corresponds to the amino acid sequence of the mature Fe_RF6318 enzyme defined in SEQ ID NO:2. The variants have improved thermal stability and/or detergent stability compared to the parent Fe_RF6318 enzyme. Preferably the substitution is V208I and more preferably the variants comprise additional amino acid changes which further increase the stability. Also disclosed are nucleic acid sequences encoding said protease variants as well as recombinant vectors and host cells for the production of the variants. The serine protease variants are applicable in laundry and dish-washing detergent compositions, in treating fibers, in treating wool, in treating hair, in treating leather, in treating food or feed, or in any application involving modification, degradation or removal of proteinaceous material.

The present invention is related to a fungal serine protease enzyme useful in modification, degradation or removal of proteinaceous material, which enzyme comprises an amino acid sequence of the mature Tr Prb1 enzyme having an amino acid sequence of SEQ ID NO: 10 or a variant thereof having similar activity.
