A method is provided for obtaining an L-amino acid or nucleic acid-producing bacterium belonging to the genuswith an optimized level of expression of the gene which influences the distribution of carbon flow, such as the sucAB genes, comprising introducing into the chromosome of the bacterium a set of in vitro constructed DNA fragments which contain regulatory elements for gene expression instead of the native elements of the regulatory region of the gene, and selecting the colonies with increased L-amino acid productivity. Also, a method is provided for producing an L-amino acid, such as L-glutamic acid, L-proline, L-arginine, L-glutamine, L-leucine, using the bacterium with an optimized level of expression of the sucAB gene.

Method for producing an L-amino acid using a bacterium with an optimized level of gene expression

L-Glutamic acid, L-proline or L-arginine is produced by culturing a bacterium belonging to the genus, which is L-isoleucine auxotrophic and has ability to produce L-glutamic acid, L-proline or L-arginine, in a medium containing L-isoleucine, to produce and accumulate L-glutamic acid, L-proline or L-arginine in a culture, and collecting L-glutamic acid, L-proline or L-arginine from the culture.

There is provided a method for producing L-leucine using bacterium belonging to the genus, which produces L-valine, L-isoleucine and L-homoleucine in an amount of less than 1% of that of L-leucine produced due to inactivation of ilvE gene coding for branched chain amino acid transaminase and produces increased amount of L-leucine due to increasing the activity of the aromatic amino acid transaminase encoded by tyrB gene.

A method for producing L-leucine, comprising the steps of: culturing a bacterium which is transformed with a DNA coding for an a-isopropylmalate synthase densensitized in feedback inhibition by L-leucine, in a culture medium to produce and accumulate L-leucine in the medium, and recovering L-leucine from the medium.
