There is provided inter alia a process for stabilizing a eukaryotic cell line which expresses PylRS and tRNAPyl and which is suitable for incorporation of a gene encoding a target protein containing one or more non-natural amino acids encoded by a nonsense codon which comprises culturing said cell line under conditions in which the adverse effect of tRNAPyl expression on cell viability and/or cell growth is reduced or eliminated.

There are provided amino acid derivatives of formula V and VI as defined herein which are pyrrolysine analogs for use in bioconjugation processes.

There is provided inter alia a DNA construct which comprises a tRNApyl coding sequence and a RNA polymerase III promoter sequence which is capable of acting to express functional tRNApyl sufficiently to support nonsense suppression in a eukaryotic expression system.

Nucleic acid molecules

There are provided pyrrolysine analogs of the formulae (X), (I), (II), (V), (VI), (VII) and (VIII), in which the a, b, d, X, Y, Z, FG, R, R, Rand Rare as defined in the claims, which are useful in bioconjugation processes and mutant proteins containing them.

The present invention relates generally to methods for identifying and using organ-specific proteins and transcripts. The present invention further provides compositions comprising organ-specific proteins and transcripts encoding the same, detection reagents for detecting such proteins and transcripts, and diagnostic panels, kits and arrays for measuring organ-specific proteins/transcripts in blood, biological tissue or other biological fluid.
