Provided herein are methods for capturing extracellular vesicles from a biological sample for quantification and/or characterization (e.g., size and/or shape discrimination) using an SP-IRIS system. Also provided herein are methods of detecting a biomarker on captured extracellular vesicles or inside the captured vesicles (e.g., intra-vesicular or intra-exosomal biomarkers).

Multiplexed phenotyping of nanovesicles

An assay for detecting gene fragments, including: amplifying gene fragments comprising single-nucleotide polymorphisms (SNPs) to form an initial amplification product; isolating single strand oligonucleotides of interest from the initial amplification product; forming a solution comprising the oligonucleotides of interest and reporter molecules, each reporter molecule having a first oligonucleotide domain configured to hybridize with a complementary oligonucleotide of interest, and a second oligonucleotide domain configured to hybridize with a complementary capture probe; hybridizing, in the solution, the oligonucleotides of interest with the reporter molecules that have complementary first domains; applying the solution to the surface of a microarray including an array of capture probes fixed to a microarray slide; capturing the oligonucleotides of interest on the microarray by hybridizing the second oligonucleotide domains of the reporter molecules with complementary capture probes of the microarray; and detecting the hybridized oligonucleotides of interest captured on the microarray.

Genotyping of mutations by combination of in-tube hybridization and universal tag-microarray

Novel copolymers provide for improved microarray performance. The copolymers are based on acrylamide, acrylate, and/or methacrylate monomer repeat units and include specific, functional monomers including a fluorinated monomer to control surface hydrophilicity such as increase water contact angle and reduce spot size when the copolymer is used in thin film form on a microarray substrate. The copolymers are prepared by free-radical polymerization and are random in nature.

Method for immobilizing biologic molecules on solid surfaces

The instant invention describes uncharged water-soluble polymers to suppress electroosmotic flow in capillary electrophoresis. The polymers in the instant invention are copolymers of various derivatives of acrylamide and methacrylamide monomers with various glycidyl group containing monomers e.g., dimethylacrylamide and allyl glycidyl ether-epoxy poly(DMA)-, copolymers of various derivatives of acrylamide and methacrylamide with various allyl group containing carbohydrates and various glycidyl group containing monomers, such as allyl &bgr;-D-pyranoside (typically &bgr;-D-glucopyranoside) or allyl &bgr;-D-furanoside, allyl glycidyl ether-epoxy poly(AG-AA) and copolymers of four different monomers including various acryl and methacrylamide, various allyl group containing carbohydrates, various glycidyl group containing monomers and various diol group containing monomers, such as acrylamide, allyl &bgr;-D-pyranoside (typically &bgr;-D-galactopyranoside or N-allylgluconamide) or allyl &bgr;-D-furanoside, allyl glycydyl ether and allyoxy-1,2 propanediol-epoxy poly(AGal-AA-APD). The subject polymers adsorb onto the capillary surface, forming a highly hydrophilic, dynamic coating that suppresses electroosmotic flow. The subject polymers may be used as a coating prior to use of an electrophoresis microchannel or may be included in a separation media contained within the capillary column. The instant coatings and media are especially suitable for applications pertaining to the electrophoretic separation of various biomolecules, such as protein and DNA.

Robust polymer coating

The present invention describes a method and apparatus utilizing a capillary coating of linear and crosslinked polymers and copolymers of acrylamidomonomers or methacrylamides bearing two hydroxyethyl residues. The method and apparatus employ this coating to maintain coating integrity and suppress electroosmotic flow under basic conditions in capillary electrophoresis.

Compounds for molecular separations

Covalently cross-linked, mixed-bed agarose-polyacrylamide matrices for electrophoresis and chromatography are disclosed. The mixed-bed matrices are formed by the copolymerization of a substituted agarose mixture with acrylamide monomers. The disclosed matrices have improved polymerization properties enabling the formation of gels which have higher porosity and elasticity than conventional gels and which are therefore particularly suited to the separation of intermediate size DNA fragments and high molecular mass proteins.
